Probiotics has been widely used in the field of livestock breeding,such aslactobacillibacteria,yeasts,enterococci and so on.Bacillus subtilis have many features of theresistance to high temperature,choline,acids and enzymes,so people paid more attention toBacillus subtilis.Therefore, we isolated and identified Bacillus subtilis from chicken feces,selected superior strain by a series of biological characteristics and evaluated its potential asprobiotics strains.1、The17strains were isolated from37chicken feces samples by conventional bacterialisolation and13/17Bacillus subtilis were identified with special PCR based on16SrRNA,which were named J1-J13.2、The results of a series of biological characteristics showed that:the average survival rate of13strains were80%,70%and50%at80℃,100℃and120℃for20min. J1,J5,J6,J8,J10were3.21×106CFU/g at PH1.5for6hours. J5,J,J9,J10,J12,J13were4.96×106CFU/g at4g/kg of choline concentration;All strains were not sensitive to mos tantibiotics. J5, J10hadsignificant inhibitory effect to pathogenic isolates after Bacillus subtilis had been cultured for24h firstly.Antibacterial effect were weak when J5, J10and pathogenic strains werecocultured at te same time. Antibacterial effect was the worst when Bacillus subtilis wereinoculated with pathogenic strains which had been cultured for24h.3、J5and J10were added to the broiler feed with1×109CFU/ml and fecals and serum werecollected regular.The results are as follows:(1)The number of total aerobic bacteria were decreased and was significant differences (P<0.05)compared with the control group at15,45and60days; The number of the totalanaerobian were increased and was significant differences(P <0.05)compared with the controlgroup at15days;The number of lactobacilli test were increased and was significantdifferences (P <0.05)compared with the control group during the entire experimental;Thenumber of fecal E.coli were decreased and the bifidobacteria was increased, they weresignificant differences (P <0.05) compared with the control group at15and30days.(2)The immunoglobulin IgA and IgG levels were increased and was significant differences(P <0.05)compared the test group with the control group at60days;The IgM,CD4+,CD8+ levels were increased but no significant differences compared the test group to the controlgroup during the entire experimental (P>0.05).(3) The triglyceride,albumin,Ca,P values were increased and was significant differences (P<0.05)compared with the control group of at45and60days;The total protein content levelswere increased and was significant differences (P <0.05) compared with the control group at60days;The total cholesterol levels was no significant differences compared with the controlgroup during the entire experimental(P>0.05).
