Study on fat deposition traits has been the focus of scholars of breeding. In recentyears, looking for candidate gene of pig fat deposition traits in order to study thepolymorphism is a common research method. However, the fat deposition traits arequantitative traits controlled by multiple genes, and regulated by the upstream gene ofthem, single gene studies cannot fully explain the genetic mechanism of the traits. Somining the genes effect fat deposition traits, to study the relationship between the geneexpression has become a key research on fat deposition. This study is based on thebiochemistry theory about pig fat synthesis, uses method of literature mining,determining the genes encoding enzymes of catalytic reaction in fat synthesis, as well asthe genes has reported to regulate these genes, to construct the lipogenic gene regulatorynetworks.This research build a regulatory network with PPARγ、LXRα、SREBP-1、ChREBPgene as the backbone. Genes encoding enzymes of fat synthesis such as FASN、ACC、DGAT、GPAT、ACSL. There are PPARγ'LXRα'SREBP-1'FASN(ACC、Thrsp、S14R)pathway, PPARγ'LXRα'ChREBP'FASN(ME、SCD1、Thrsp、ACC、FASN、S14R)pathway and any others. FASN、ACC、DGAT、GPAT、ACSL catalytic enzyme genesare in the downstream pathway.Information of catalytic enzyme genes in pig fatty acid chain elongation is less,relatively. The chromosome localization results show these enzyme genes are located inthe chromosome that adipose synthesis genes distribution is relatively concentrated. Sowe can go to research further to mine marker genes for molecular breeding.Bioinformatic analysis of fat synthesis related gene FASN, ACACA, Thrsp, ACSL1,predicted primary structure, secondary structure, signal peptide, transmembrane structure,subcellular localization, structural domains of their encoded proteins, to provide areference basis for further study.By qPCR on the expression of ACACA, Thrsp, ACSL1gene at the transcriptionallevel analysis, expression of three genes showed a high consistency, and the three geneexpression correlation between reached significant or extremely significant level, which shows there is relationship between the expression of certain regulation of three genes.The correlation is the basis for study on the mechanism of regulation of transcription, wecan continue with a series of studies, promoter region, transcription factor binding sites,Combinations of DNA and transcription factors, to determine the transcriptionalregulation mechanism in detail.
