Brown planthopper[Niaparvata lugens (Stal)], one of the major pests for rice in China and many other Asian countries producing rice, can cause damage through long-distance migration, and disasters are easy to outbreak if conditions suit. Insulin is an important kind of endocrine hormone, which plays a vital role in regulating the physiological and biochemical reactions for the livings, thus it is the potential target for pest control. This paper mainly researches on Insulin-like peptide(ILP), and the following results are obtained:(1) We successfully cloned4insulin-like genes of brown planthopper, Nlllpl-4. Through alignment of sequences, it is shown that the structures characteristics of MlILP1and MILP3are more similar to the insulin-like peptide, and NlIlp2is more similar to the insulin-like growth factor.(2) Through the qRT-PCR technology, we analyzed the space-time expression patterns of brown planthopper’s Nlllp1-4.(3) Through the RNA interference (RNAi) technology, we find that silent NlIlp2can shorten the growth time of brown planthopper from3instar nymph to adult, and silent MlILP3prolong the growth time.(4) It is found that silent MlILP1can reduce the weight of female adult brown planthopper, MlILP-4can increase the content of triglyceride in female adult brown planthopper, silent MlILP1and MlILP3can increase the content of glucose in female adult brown planthopper, and silent Nlllp3can reduce the content of glycogen in female adult brown planthopper.(5) The polyclonal antibody against MlILP3was successfully prepared. The research on insulin-like gene of brown planthopper in this paper has important significance for pest control and a deeper understanding of insulin’s biological functions.
