The Preparation Of Eucommia Ulmoides Injection And The Bacteriostasis Effect Of Pathogenic Bacteria

In order to studied the preparation of Eucommia ulmoides injection and the bacteriostasis effect ofmajor pathogenic bacteria causing mastitis.This experiment adopted the single factor method todetermine the optimum range of medicinal additives and pH, through the L9(33)orthogonalexperiment method to determine the best dosage of medicinal additives and pH value. Using singlefactor experiment to determine the medicinal dosage of activated carbon,adsorption temperature andadsorption time, select the best adsorption conditions. According to the veterinary drug standard ofthe People’s Republic of China (2010edition),checked the traits, visible foreign matter, relatedsubstances, identification test and the contant of Chlorogenic acidof Eucommia ulmoides injection,and to evaluate its security and stability. Through the methods of tube double dilution and drilling tostudied the bacteriostasis effect of E. coli, staphylococcus aureus, Streptococcus uberis andStreptococcus dysgalactiae.The results show that the optimum range of medicinal additives and pH of Eucommia ulmoidesinjection was sodium bisulfite (W/V)0.14%～0.16%, tween-80(V/V)0.05%～0.06%, the optimumpH range was5～6, and the best dosage of sodium bisulfite was0.14%,0.05%tween-80, the pHvalue of5. The best preparation process was added0.14%sodium bisulfite,0.05%twain-80andpH=5to the pretreatment of extract of Eucommia ulmoides,0.2%medicinal carbon (W/V),45℃adsorption25min,0.45μm microporous membrane coarse filter,0.22μm microporous membrane finefiltration, packaging, potting. Quality test results showed that the Eucommia ulmoides injection wasclear and transparent brown liquid, had no visible foreign matter, proteins, tannins and resins, andother related substances, the content of extract of Eucommia ulmoides was1g/mL and activeingredient of chlorogenic acid not less than8mg/mL. Conforms to safety tests of the veterinary drugstandard of the People’s Republic of China (2010edition) of pyrogen inspection, abnormal toxicitytest, allergy and hemolysis test. The high light test, hyperthermia test and high humidity test resultsshowed that light and hyperthermia caused the content of chlorogenic acid lower of injection, shouldbe kept away from light and at room temperature. Accelerated test and long-term test results showthat eucommia ulmoides injection quality is stable, no visible foreign matter or related material,conform to the veterinary drug standard of the People’s Republic of China (2010edition) stability teststandards. Eucommia ulmoides injection have high sensitivity to of Staphylococcus aureus, and sensitivity to E. coli, Streptococcus uberis and Streptococcus dysgalactiae. The MIC were0.125g/mL, the MBC of Staphylococcus aureus was0.125g/mL, others was0.25g/mL, the bacteriostaticring size respectively was18.42mm,14.21mm,12.01mm and12.89mm.In summary, the Eucommia ulmoides injection preparation process reliable, security and stable,controllable quality, have a good bacteriostasis effect of four kinds of pathogenic bacteria causingmastitis.
