The expression of nutrient transporter genes GLUT2, GLUTS and SGLT1 directly affect the intestinal absorption of nutrition function. In order to reveal the transcription factor GATA2 regulate the expression and regulation mechanism, which these three kinds of function gene in the molecular level. The content of the experiment is the influence of the over expression of the GATA2 transcription factor to the three functional gene.Firstly, According to the bioinformatics software of ModuleMasterl.4、http://tfbind.hgc.jp indicated that it plays a role in regulation sites the expression of monosaccharide transporter gene GLUT2, GLUTS and SGLT1. Secondly, the eukaryotic expression vector pcDNA3.1-GATA2 has been constructed and transfect into chicken intestinal epithelial cells by liposome mediated method. The expression of GATA2 and absolute monosaccharide transporter gene GLUT2, SGLT1 and GLUTS content were detected by real-time fluorescence quantitative PCR.. The result suggests that the expression of mRNA in the transfected group is six times higher than the empty vector group and blank cells group.From the statistical analysis, we can see the transfected group showed significant difference (P< 0.01). Among the pcDNA3.1-GATA2 transfected primary chicken intestinal epithelial cells, the empty vector group and blank cells group (non transfection group), which proved that the GATA2 transcription factor has a success. The results suggest that the expression of the GLUT2 showed significant difference (P< 0.01). Among the pcDNA3.1-GATA2 transfected primary chicken intestinal epithelial cells, the empty vector group and blank cells group (non transfection group), which proved that the GATA2 transcription factor has a negative role in the regulation of monosaccharide transporter gene GLUT2. The expression of transfected group and empty vector group, blank cell groups in SGLT1 and GLUTS were not significant difference (P> 0.05), The results will help to understand the biological functions of GATA2 in the chicken nutrient absorption and select suitable transcription factor bindingsite provide a theoretical basis.
