[Objective] Tibial dyschondroplasia (TD) can lead to thickenness of tibial growth plate, and accumulation of large chondrocytes. It is characterized by the presence of a non-vascularized and unmineralized cartilage in the growth plate. Differentially expressed genes about occurrence and development of tumor were selected through gene microarray including HDAC1, MTA1, H4, PCNA and so on. To explore the relationship between these genes and TD in chicken, this experiment was conducted to research the expression of these four genes.[Methods] This experiment used the Real-time qPCR, immunohistochemical technique to research expression changes of HDAC1、MTA1、H4 and PCNA genes in quantitative and protein positioning by constructing TD model in broiler chichen at the early stage.[Results] Real-time qPCR results showed that, Compared with the control group, HDAC1, MTA1, H4, PCNA in the growth plate of TD model had shown the differential expression significantly in transcriptional level. The expression of HDAC1 was up-regulated and had significantly differential expression on days 1,2,4 and 6 (P<0.01); The expression of MTA1 was found up-regulated and had significantly differentiated expression on days 1 and 2 (P<0.01); The expression of H4 was up-regulated and with significantly differential expression on days 2 and 4 (P<0.01); The expression of PCNA was down-regulated and had shown a significant differential expression on days 1,2 and 4 (P<0.01).(2) Immunohistochemistry results showed that, HDAC1 and MTAl protein had different levels of expression in tibial growth plate chondrocytes at different zones in TD model. During the trial, HDACl protein were expressed all over in different zones, which in the proliferative zone on day 1 and the hypertrophic zone on day 6, chondrocytes showed highly significant difference (P<0.01), and the proliferative zone on day 2 and the hypertrophic zone on day 1 and 2 were showed significant differences (P<0.05); MTA1 protein expression were observed all over on day 1,2 and 6 and showed highly significant differences (P<0.01) in all zones, except the prehypertrophic zone on day 2 with no difference expression.[Conclusion] In the mRNA level, HDAC1, MTA1, H4 and PCNA had different expression in tibial growth plate of chicken TD model which was the same with gene microarray before. In the translational level, HDAC1, MTA1 also had a significant difference which indicated that differential expression at protein level was related to the abnormal proliferation of chondrocyte in tibial growth plate of TD model.
