In this experiment, the inducing conditions and growing conditions of adventious roots and hairy roots of Sophora flavescens Ait.,the conditional optimality were studied. The main results were as follows:(1) Sophora flavescens Ait. aseptic seedlings,in the 1/2MS (half of macroelement solid)medium without hormone,dark culture for a week,then growth the light. That was the highest germination rate. To provide for the explant material in the experiment.(2) The cotyledon, leaves, stems, roots of Sophora flavescens Ait. as explant, into the B5 medium and MS medium.The best explant was root.And the best medium was B5.In B5 medium, explants can differentiate of a lot of adventitious root rapidly.Then mixed with different levels of IBA or NAA.According to this experiment,Sophora flavescens Ait. adventious roots had the most vigorous growth and maximum biomass when concertration of IBA was 1.5mg/L.It had the maximum content of matrine.(3) The cotyledon,leaves,stems,roots of Sophora flavescens Ait. as explant, into the different medium could be induced callus.The best explant was cotyledon.Both 2,4-D and 6-BA have great function in promoting induction and growth of Sophora flavescens Ait. callus. Different explants could induce callus in MS+1.5mg/L of 2,4-D+1.0 mg/L 6-BA.The cotyledon,stem, and root had a high callus reduction rate about 98%,and had a lower rate about 60% of leaves.(4) The cotyledon, cotyledonary node, leaves, stems, roots of Sophora flavescens Ait. as explant,infescted by R1000,R1601,15834,A4 and LBA9402 with inject.According to this experiment, R1601 and cotyledonary node was the best condition.The inducting rate was 33%. It can observe that the hairy root will groe by itself and biomass would increase.The color of hairy root would changed darken with the extende cultirate time.The inductivity can be improved to 50% or more if add a certain amount of AS to the present mrdium.
