Study On The Developmental Expression Of ChREBP Gene In Three Tissues Of Mashen And Large White Pigs

ChREBP which is a transcription factor binds to the promoters of glycolytic and lipogenic enzyme genes to regulate its transcription. This study detected the expression of ChREBP mRNA and protein in liver, longissimus dorsi and backfat at seven stages (1-,30-,60-,90-,120-,150-,180- day old) of Mashen and Large White pigs by qRT-PCR and Western blotting. The main results were as follows:1. The expression profile of ChREBP mRNA in liver and longissimus dorsi of two breeds was like"wave". The highest expression in liver of Mashen and Large White pigs was at 30- and 60- day, respectively. That at 1- day was the lowest in longissimus dorsi of two breeds, the expression peak was at 60- and 90- day, respectively. The highest expression in backfat of both breeds was at later stages of growth. Comparision between breeds, the expression of Mashen pigs was significantly higher than Large White at 30-,90-,150- and 180- day in liver and the whole stages in longissimus dorsi, while there was opposite result at the other stages in liver and the whole stages in backfat. Comparision among tissues, the expression in liver of Mashen pigs was highest, followed by the longissimus dorsi and backfat, successively; that in longissimus dorsi of Large White pigs was the lowest, and the tissues which had the highest expression were in liver and backfat at early and later stages of growth, respectively.2. The highest expression of ChREBP protein in liver was at 90- day of Mashen pigs and 60- day of Large White, respectively. The expression at 1- day was the lowest phase in longissimus dorsi of two breeds, and it had a rapid rise afterwards. While that at 30- day was the higtest in backfat. Comparision between breeds, the expression of ChREBP protein in liver and backfat of Mashen pigs was lower than Large White. That in longissimus dorsi of Mashen pigs was higher than Large White except for 90- day. Comparision among tissues, the expression in longissimus dorsi of two breeds was significantly higher than the others, except for 1- and 60 d of Large White pigs.The results indicated that ChREBP mRNA and protein expression had significant differences in different breeds, tissues and days of age, it suggests that ChREBP plays an important role in the regulation of body fat deposition of pigs.
