Sterol regulatory element binding proteins(SREBPs) is a significant kind of nuclear transcription factor.Among them,SREBP1c can combine with the sterol response element(SRE) of acetyl-CoA carboxylase(ACCa),fatty acid synyhase(FAS) and stearoyl-CoA desasturase1(SCD1) gene and activate the transcription of the de novo fatty acid synthesis enzymes.Which can regulate the de novo fatty acid,and then affect the lipid synthesis.This study detected the expression of SREBP1c and SCD1 mRNA in liver,longsissimus dorsi and back fat.At the same time,SCD1 was detected the expression of protein in liver,longsissimus dorsi and back fat by using Western blotting.The results were as follow:1.The expression profile of SREBP1c mRNA in three tissues of two breeds was similar.In liver,the highest expression of Mashen and Large White pigs was at the age of 120- and 180-day in fattening period respectively.However,the expression of Mashen and Large White pigs was lower at the age of 150-day.In back fat tissue,the highest expression of Mashen and Large White pigs was at the age of 30- and 150-day respectively.The expression of two breeds was significant difference in the same period(besides 90-day).In longissimus dorsi tissue,the expression of two breeds was low in the same age except that 90-day of Mashen and 180-day of Large White was higher.In three tissues,the overall expression profile of SREBPlc mRNA in Large White pig was back fat>liver>longissimus dorsi,the expression of Mashen was higher in longissimus dorsi in the same period(besides 120-day).2.Besides the expressional pattern of SCD1 mRNA in Large White of back fat was like " stepped up",The other tissues in two breeds were similar.The peak expression of three tissues in two breeds is 180-day.In liver tissue,the expression of Mashen and Large White have similar trend,the expression of SCD1 mRNA is extremely significant difference at the same age of two breeds.In back fat tissue,Mashen and Large White pigs performed the expression of progressive increasing,the expression of Mashen was higher than Large White pig in two breeds of the same age(besides 60-day).In longissimus dorsi tissue,the expression of Mashen and Large White was extremely significant difference at the age of 60-day,the others were significant difference or extremely significant difference,the expression of Mashen was higher than Large White in later period of growth and development.In three tissues,the overall expression of back fat in two breeds was advantage tissue,the peak of expression was 180-day in two breed of three tissues.3.The expression profile of SCD1 protein in three tissues of two breeds was similar. In liver tissue, the highest expression of Mashen and Large White pigs was at the age of 180-day; In back fat tissue,on the whole,the SCD1 protein expression of Mashen was higher than Large White in the same age; In longissimus dorsi tissue, the highest SCD1 protein expression of Mashen and Large White pigs was at the age of 150-and 30-day,the expression of Mashen was significantly higher than Large White in the later period of growth and development;In three tissues,in the later period of growth and development,the expression of Large White in liver was significantly higher than the other tissues at the same age,the expression of Mashen in three tissues did not have an obvious regularity.
