The genetic polymorphism of five candidate genes in Hotan sheep keyatin family are studied through PCR-SSCP and DNA sequencing techniques in the test,least-squares method in SAS8.1 is usedtoanalyse the relationship betweenwool length and fineness,and polygenic combination’s influence on Hotan sheep wool traits is further analyzed. This article is aimed to find out the genes related to wool traits, and improve Hotan sheep wool traits, and provide scientific basis for breeding, thereby, increase the income of herdsmen.KAP6.1 gene occurstwo mutations at C51 T and G57 A in the nucleotide sequence of the non-coding region intron; difference ofcorrelation resultsbetween the gene and the wool length and fineness are not significant(P> 0.05).KAP7 gene mutationsoccurrsin the nucleotide sequence T110 G and G268 A place. The difference of wool length between AA and AB genotype isnot significant, but the difference between AA, ABgenotype and BB genotype issignificantly; three genotypes’ influence ontraits of the wool finenessisnot significant. So KAP7 gene may serve as a DNA moleculemarkthat can represent Hotansheep wool length.KRT27 genemutates at thelocation of G49 A, T58 Cin nucleotide sequence; comparing SS genottpe withST and TT genotypes, their wool length and fineness are different significantly(0.01<P<0.05); comparing ST genottpe with TT genotype, wool length and fineness differences arehighly significant(P<0.01). KRT27 gene may serve asa DNA molecule markrepresentingHotansheep wool length and fineness.KRT31 gene mutates at C52 Tin nucleotide sequence. With the correlationanalysis of wool lengthtraits, differencebetweenEE genotype and EF, FF genotype is significant(0.01<P<0.05), wool length of FF genotype was significantly longer than the EF genotype(P<0.01); with the correlation analysis of wool fineness traits, differences between the three genotypes are not significantly different(P>0.05). KRT31 gene may be labeled as a DNA molecule representing Hetman sheep wool length.The five genesis excrement, KAP8 gene polymorphism was not detected; KAP6.1, KAP7, KRT27 and KRT31 in Hogan sheep population’s mutation sites are all in Hardy-Weinberg equilibrium state(P>0.05).In Hogan sheep populations, using genetic polymerization techniques to analysis, the results show thatheterosis effects not reflected on the wool fineness after the combination of advantage genotypes, while wool length showed a corresponding advantage. It provides corresponding reference basis for further use of multi-gene pyramiding breeding technology and research on sheep wool traits in the future.
