Yangtze River Delta white goat is the only goat variety which is not only characterized by its high quality brush hair(namely Type Ⅲ Hair), but also raised for meat, derma and wool, the Type Ⅲ Hair is specialized for top-grade writing brush. Yangtze River Delta White Goat is one of the precious and irreplaceable germplasm resources, because of its unique performance on production of Type Ⅲ Hair, it was also named as writing brush goat. At present, study on high quality writing brush hair traits of the Yangtze River Delta White Goat main at the macro, the research has not been carried out in the molecular level. This study that based on studies that previous relevant molecular mechanisms of our group, do further studies about high quality writing brush hair traits of the Yangtze River Delta White Goat by RNA-Seq technology to further explore the molecular mechanism of the formation of this trait.Through the study, high-throughput sequencing of the transcriptomes from the high quality individuals and non-high quality ones of Yangtze River Delta White Goat was carried out to identify important genes and related pathways that may influence the traits of hair quality,and polymorphism analysis of related genes was also executed by PCR-SSCP. At the same time, follicles structures of high quality individuals and non-high quality ones were compared by frozen section technique, in order to find the differences between the two characters of the follicles structure.The results showed that:(1) there are 295 genes that expressed differentially between the non-and high quality individuals, among which there are 132 up-regulated genes and 62 down-regulated genes,101 genes expressed specifically in high quality individuals. (2) the results of GO and Metabolic Pathway Significant enrichment analysis of the differential genes indicate that MAP3K1 gene and MAPK signal path may play important roles in the formation of high quality writing brush hair traits. (3) 10 SNP locus of MAP3K1 gene reside in neither high quality nor non-individuals according to NCBI. (4) there are no difference of follicles structures between the high quality and non-individuals from the dorsal neck and back dermas, but primary follicles density in the dorsal neck dermas of the high quality and non-individuals is significantly lower than the density in back dermas of the high quality individuals.In this study, annotation of Unigenes from both of the phenotypes had been obtained, differential expression gene pool with all the differentially expressed genes of the metabolic pathways was established, which can provide theoretical evidence for further study of molecular mechanisms on the process of Type III Hair traits in Yangtze River Delta White Goat.
