The Yangtze river delta white goat is the only goat breed that produces high-quality brush hair,which is specifically used in top-grade writing brushes.In the production process,the classification of brush hair is mainly depends on the experience,there are no detailed statistical results for the physical parameters of the brush hair,therefore,it is necessary for a set of classification standards for the processing of brush hair.In this study,the length,fineness,strength and scale of the brush hair were measured,these data can provide a certain data support and scientific theoretical basis for the industry to formulate the classification standard of brush hair.Yangtze River Delta White Goat is a unique precious variety in China,and there has been little research,especially molecular research,on the traits that result in high-quality brush hair in the Yangtze River Delta White Goat.Previous studies have demonstrated that the CMTM3 and DUSP1 genes are involved in the growth cycle of high-quality brush hair,and the formation of high-quality brush hair traits.The relationship of CMTM3 and DUSP1 methylation with these brush hair traits were explored using the bisulfite sequencing method.This study results showed that,1.In the present brush hair market,type ?includes the goat tail,goat beard,Shangzhua/Cuzhuafeng,type ? includes the Baijianfeng,Xichangfeng,Laoguangfeng,Jiaozhuafeng,type ? includes the Zhongguangfeng,Changguangfeng,Duanguangfeng,Xiguangfeng,Cuguangfeng.And the type I hair is medullatry wool,its range of fineness varies greatly,usually between 50?120 ?m,and the scales are non circular scales.The type ? hair is medullatry-unmedullary wool,its fineness usually between 80?120 ?m,and the scales are mostly rectangular.Type ? hair is unmedullary hair,its fineness usually between 70?90 ?m,and the scales are mostly ring-shaped or fish scale.2.The results indicated that the relative expression levels of the CMTM3 and DUSP1 genes were higher in non-high-quality brush hair than in high-quality brush hair.Between 2000 bp upstream and 200 bp downstream of the transcription start site,the CMTM3 gene has two CpG islands,whereas DUSP1 has a single CpG island.Furthermore,the CpG sites of the DUSP1 gene were not methylated,and the methylation level of CMTM3 was negatively correlated with the gene expression level.3.In this study,we found that the CpG sites of the DUSP1 gene were not methylated.And we suspect that the DUSP1 gene regulates the formation of high-quality brush hair via its non-methylation,which results in changes in its expression,thereby affecting the expression of upstream and downstream gene changes,which in turn indirectly affects the formation of high-quality brush hair traits.4.The results of androgen test showed that the level of androgen in the high quality brush hair group was significantly higher than that in the non high-quality brush hair group,ewe group and castrated ram group.5.The CMTM3 gene is methylated,resulting in its decreased expression,thereby causing AR activity and the level of androgen to increase;the high level of androgen promotes the growth of high-quality brush hair.
