Strawberry "JC" which taken from Japan, planted in many parts of the large area and rich in nutritional value, has wide adaptability, broad market, is worth for high economic, and is suitable for planted. But the traditional way of breeding cycles long, lead to low propagation coefficient, which degraded seedlings quickly.To provide the technical support and the guarantee to realize the industrialization, this study based on the tissue culture, with the stem tip of stolon as explant, explored the influences to the rapid propagation and the transplanting domestication of strawberry "JC" and then analyze the quality of fruit. The results showed that:1. The seedlings on the MS medium got large proliferation and grew green leaves; BA, as cytokinins, which proliferated more than that of TDZ, KT with 1.2 mg·L-1; NAA with 0.10 mg·L-1, is more benefit for propagation, in which the seedlings got large fresh weight, dry weight and high propagation coefficient;There was no significant difference between the sucrose and powered sugar with 30 g·L-1, which is obviously higher than other treatments; with the growth of illumination intensity which is more suitable for the proliferation of strawberry at 1600 lx, the seedlings increased firstly and then decreased with increasing light intensity; So the most suitable medium of "JC" for proliferation which up to 12.4 was sucrose at 30 g·L-1, with BA 1.2 mg·L-1, NAA 0.10 mg·L-1 and the illumination intensity at 1600 lx, It can be used for Industrial production to reduce the cost of sugar instead of sucrose.2. The seedlings got the largest rooting rate on 1/2 MS culture medium; IB A was the best for rooting on growth regulator levels among IAA, IBA, NAA, the root rate was as high as 100%, and the weight, dry weight and the number of root up to a maximum, respectively was 75.4 mg,8.4 mg,16.7 on rooting with 0.5 mg·L-1, which brought the most and the thickest roots at length of 2.4 cm. Activated carbon could inhibit the formation of phenolics and promote the increase of the root of strawberry. The growth vigor of strawberry "JC" increased and then dropped, with the increase of concentration of activated charcoal, with 1.0 mg·L-1, which produced the stronger and more fibrous root and the most exuberant plants, significantly better than others;The sucrose is suitable for the root of strawberry "JC" with 30 g·L-1 which produced the largest fresh weight, dry weight, and root number, were 87.6 mg,8.6 mg,18.1, significantly better than other levels.3. The survival rate of perlite and peatmoss mixxed was lower than that of vermiculite and perlite composite, which had the highest survival rate and rooting rate at 98.2%、96.3% when their proportion was 1:1.IBA was the better auxin than IAA during the outside bottles rooting.The concentration of IBA, with 0.7 mg·L-1, which produced the more fibrous and stronger longer root and the most exuberant plants, it also reach the highest survival rate, significantly better than others. The two ways of strawberry seedlings, the tube seedlings and the somaclone outside of bottles could be normal to bloom and fruit after domestication.4. The results of the fruit domesticated by tube seedlings showed that, the treatment of T2 got the highest content of reducing sugar、Vc and Phenolic content, was 564.1 mg·g-1,0.639 mg·g-1,0.029%, significantly better than other levels.
