| This study used blueberry varieties such as Duke,Spartan,Northsky,and Sunshine Blue as test materials.To explore the photosynthesis characteristics of different varieties of blueberries,the hormone concentration of induced primary culture,the suitable medium for rooting in tissue culture seedling bottles,the in vitro and ex vitro rooting for the test-tube,and the nutrient solution formula of blueberry tissue culture seedlings after domestication and transplanting.1.Before blueberry tissue culture,different varieties of blueberry explants were cultivated,and the growth characteristics and photosynthetic characteristics of each variety were studied.The conclusion is:the Bluecrop leaf color index is the highest,followed by Misty;the Sharpblue leaf area is the largest,the Sunshine Blue is the second;the main stem in the Northsky is the thickest,the Sunshine Blue;the Duke plant is the highest,the Northsky is the second;the Northsky number of new branches is the highest,followed by Misty.The main photosynthetic parameters of Duke,Spartan,Sunshine Blue,Sharpblue and Northsky were determined.The results showed that different varieties had different utilization ability for photosynthetically active radiation,among which the northern sky was the strongest and the sharp blue was the weakest.Duke,Spartan,and Sunshine Blue are in the middle;photosynthetically active radiation is above 800μmol·m-2·s-1,which is beneficial to photosynthesis of blueberries;photosynthetically active radiation is 1200μmol·m-2·s-1,and 5 varieties are net.The photosynthesis rate reached the maximum value;the order of transpiration between varieties was:Northsky>Sunshine Blue,Spartan,Sharpblue>Duke.2.Using stem segments as explants,suitable medium for the induction of primary cultures of four varieties of Spartan,Duke,Sunshine Blue and Northsky.The results showed that the medium suitable for differentiation of stem segments was added with different concentrations of ZT in modified WPM.The suitable ZT concentration of Sunshine Blue and Spartan was 1.2mg/L,the Northsky ZT was 0.8 mg/L,and the Duke was ZT0.5 mg/L.3.The suitable medium for rooting of four varieties of blueberry tissue culture seedlings in Spartan,Duke,Sunshine Blue and Northsky was studied.The conclusion is that the influence of the components of the medium on the rooting rate is from the deep to the shallow order:IBA concentration>sucrose and agar concentration>basic medium.The most suitable rooting medium was 1/2 WPM+IBA 0.5 mg/L+sucrose 15 g/L+agar 8 g/L.4.Comparative study on growth and development of blueberry seedlings in vitro and ex vitro rooting for the test-tube.The results showed that there were no significant differences at net photosynthetic rate,transpiration rate,intercellular CO2 concentration and stomatal conductance.There were also no significant difference at water use efficiency,chlorophyll content,soluble sugar content and soluble protein content.There were no significant differences at growth indicators including main stem diameter,number of new shoots,main stem length and leaf color index.Both of these methods can be used for blueberry reproduction.Considering cost saving and increasing breeding speed,the ex vitro rooting is an effective method for the test-tube seedlings of blueberry.5.Study on the nutrient solution of blueberry tissue culture seedlings after domestication and transplanting.The blueberry tissue culture seedlings after domestication and transplanting are watered with a large amount of nutrient solution and different nutrient solution for seedling cultivation.The fresh weight and dry weight,fresh shoots and leaves,shoot growth,root activity,chlorophyll content,carotenoid content,SOD activity and other indicators of blueberry seedlings cultivated by each nutrient solution were determined and analyzed by membership function.Data,the results determine the optimal nutrient solution of nutrient solution is NO3-0.8155 mmol/L、NH4+1.6187 mmol/L、P 0.2082 mmol/L、K 2.1021 mmol/L、Ca 0.9699mmol/L、Mg 0.2502 mmol/L、S 1.5988 mmol/L。... |
