| With the initial completion of the bovine genome project and the development of molecular techniques, it becomes possible to use of molecular markers for marker-assisted selection (MAS), where one of the most hot molecular markers is the single nucleotide polymorphism (SNP). (Two of the main goals as beef cattle breeding to beef cattle breeders are the carcass traits and the meat quality traits of beef cattle in good condition as symbols). In this study, using bioinformatics and molecular biology, the correlation analysis of relevant data were performed between 12SNPs of the UCP3ã€THRSP genes and 9 traits in 65 cattles. The mRNA expression of tow genes were determined using real time PCR at different marbling score and the results were as the following:1.Altogether 5 SNPs in UCP3 gene were detected using PCR-RFLP. The association between SNPs and carcass traits and meat quality traits was analyzed with general linear model, and the result shows that:Altogether 5 SNPs were detected in intro 5 of ucp3 gene.(1) The C786G was associated with fat color grades and the fat content, the mean of genotype CG was significantly higher than CC genotype (P<0.05); The content of fresh water of animals with CC genotype was higher than those with CG genotype (P<0.05).(2) The C782G was associated with fat color grades and the fat content, the mean of genotype CG was significantly higher than CC genotype (P<0.05); The content of fresh water of animals with CC genotype was higher than those with CG genotype (P<0.05).(3) The A559G was associated with fat color grades and the fat content, the mean of genotype AG was significantly higher than AA genotype (P<0.05); The content of fresh water of animals with AA genotype was higher than those with AG genotype (P<0.05).(4) The site of C508T was not significantly associated with any carcass traits and meat quality traits (P>0.05).(5) The animals with genotype GT in the site G457T is significantly higher than those with genotype GG in weight before slauteringã€eye muscle lengthã€triangle belly beef grade% backfat thickness% fat color gradeã€meat color gradeã€beef marbling grade. The content of fresh water of animals with GG genotype is higher than those with GT genotype.2.7 SNP sites were detected in THRSP gene by sequencing and the PCR-RFLP method. The correlation analysis is performed between 7 SNPs and carcass traits and meat quality traits. The results are as follows: There are 6 SNP in intron 1 of THRSP gene.(1) In intron 1, the site of A790C, A724G, T682C, T470C, G378A were not significantlyassociated with any carcass traits and meat quality traits.(2) The animals with TC genotype in the site of T448C in Intron 1 are of significantly higher marbling grade than those with TT genotype (P<0.05).(3) Extron 1 of THRSP contains one SNP site A193G. The genotype of AA in this site was associated with significantly higher fresh meat water content (P<0.05) and AG was associated with higher fat content (P<0.05).3. There is no significant difference between the UCP3 expressions in animals with significantly different marbling grade (P>0.05). The THRSP expression in animals with higher marbling grade is significantly higher than those with lower grade (P<0.05). |
