The peony is the specific flower in our country. However the traditional modes of reproduction have the problems of low propagation coefficient, long breeding cycle and so on. That has made the cultivation and production of peony is difficult to form large-scale and can’t meet the needs of markets. The plant tissue culture technology is one of the effective means for rapid propagation of the peony, which not only can shorten cultivation period, increase cultivation index, produce on a large scale, but also keep the superior character of the mother plant. Somatic embryogenesis regeneration plants as an important plant regeneration method in tissue culture, which has been successful in many woody plants. The researches on peony’s somatic embryogenesis are rare relatively at present. In this study, I use the scaly buds of Paeonia suffruticosa ‘Fengdanbai’ as explants in vitro culture. Firstly, make sure the most suitable medium and cultivation quomodo for the early somatic embryo induction of the peony through the study of different exogenous hormones, metallic silver ions and cultivation mode influence on the induction of somatic embryos of peony. Secondly, study the induction of somatic embryos of peony and the changes of enzyme(POD, SOD, CAT and APX) activity, soluble protein content, endogenous hormone content and balance of endogenous hormones in the most suitable way. Do the observation of tissue section at the same time. Finally, observed the effect in the process of the early embryogenesis synchronization of peony through the paraffin sections. In order to offer some theory and technology to establish the stable and efficient reference system, I explored the feasible conditions of the early somatic embryogenesis of peony in this experiment. The main results were as follows: 1. Peony callus as material, studied the different exogenous hormones, metallic silver ions and cultivation mode influence on the induction of somatic embryos of peony. The results showed that: The added picloram(PIC) treatment under different culture mode induced the most calli, and the appearance of them accorded with the characteristics of embryogenic callus. 2. Peony callus as material, studied the added PIC treatment on the induction of the early somatic embryos of situation of peony. The results showed that: in the 18 D, callus inducing culture began to form embryogenic cell, 18-30 d formed a large number of embryogenic cell, the 30 d appeared early globular embryo, and the 36 d was observed the heart-shaped embryo. At the same time, in the process of the formation of embryogenic callus, found the existence of non embryogenic cell, embryonic cell and early globular embryo coexistence. 3. Peony callus as material, studied the changes of antioxidant enzyme activity in the process of the early somatic embryogenesis with the added PIC treatment. The results showed that: 0-30 d the POD activity increased rapidly. In the 30 d the globular embryo formation stage, it reached a maximum value, then decreased slightly, but still maintained at a higher level. 0-18 d the SOD activity(embryogenic callus stage) increased rapidly and remained at a high level, the 24-30 d dropped rapidly and appeared the minimum value on the 30d(globular embryo period), and then quickly rebounded. The changing of CAT activity is the same trend with POD activity. 0-24 d increased slowly, 24-30d(globular embryo period) rose rapidly then reached to the highest value, 30-36 d dropped sharply, then changed slowly and remained at a high level. 0-30 d the APX activity decreased slowly and in a small decline then began to recovery quickly. 4. Peony callus as material, studied the changes of the soluble protein content in the process of the early somatic embryogenesis with the added PIC treatment. The results showed that: 0-30 d the rate of protein synthesis was accelerated markedly, which reaching the maximum value in the 30 d and then its content began to decline. 5. Peony callus as material, studied the changes of the endogenous hormones content and the balance between them in the process of the early somatic embryogenesis with the added PIC treatment. The results showed that: 0-30 d IAA content increased and in the 30 d that is the globular embryos formation period which reached the maximum value. Then it began to decline, but still maintained at a higher level. GA3 content showed a declining trend in the early stage of the treatment, but in a little rangeability, and then got to the minimum value in the globular embryo and the heart-shaped embryo stage. The changing of ABA content is similar to the IAA content. At the early stage of the treatment, ABA content processing increased. In the 30d(globular embryo formation stage) reached the maximum value then began to decline. At the each stage of the peony somatic embryogenesis, the mass ratio of ABA, IAA and GA3 showed dynamic changes, and the ratio of ABA / IAA and ABA / GA3 would increased in the mid and later of treatment. 6. Peony callus as material, study the different treatments on the synchronization of the early somatic embryogenesis. The results will be obtained in about May 10, 2015.
