| With red and juicy delicious flesh, strawberry has rich fruit aroma and high nutritional value, as well as the efficacy of consolidating the gums, freshing breath, moisting throat, and also can promote gastrointestinal peristalsis, help digestion, ease constipation, prevent hemorrhoids and colon cancer. Therefore it has been widely popular with consumers. Strawberry cultivation in Shanghai has developed rapidly in recent years, while asexual reproduction of strawberry susceptible to infected virus in the production process of breeding seedlings. According to statistics, strawberry sexual degradation generally caused 30%- 80%production reduction due to virus infection. Since chemical drugs has little effect on virus disease prevention and potentially causes serious pesticide pollution, so the production of more virus-free seedling is used to control the spread of the virus disease. Stem tip culture is one of important way to obtain virus-free strawberry plants. This paper briefly describes strawberry production situation of the Shanghai region as well as at home and abroad. The high quality varieties, Benihope Strawberry, was selected by field experiments and its stem tip seedling cultivation technology was conducted a series of research. Then the contrast test of virus-free seedlings and non-virus-free seedlings obtained the following main results.1, After comparing plant characteristics, fruit quality and yield of several main varieties in Shanghai Qingpu District and several varieties in Zhejiang province respectively,, we selected the cultivars red cheek strawberry with high fruit quality and yield and more suitable for the local climate.2, survival and growth of stem tip of Benihope Strawberry required the participation of 6- BA, when inducing stem tip shoot differentiation, which with no requirement for IBA concentrations, and the optimum medium is MS + 6-BA1.0 mg/L; The optimal medium for transgenerational is MS+6-BA0.5mg/l+IBA0.05 mg/l, and the value-added coefficient is 5.38; Regeneration shoots transferred to 1/2MS + IBA 0.1 mg/L rooting best with 100% rooting rate, the average root number 5.24 per strains.3) Comparative tests were carried out in the field of non-virus-free virus-free seedlings, which were divided into numerous trials and cultivation trials. Virus-free seedlings exhibited obvious advantages of non-virus-free seedling, and when breeding seedlings, they took root more quickly, had more stout stem, more vigorous growth, more strong resistance, virtually had disease and generally didn’t need pesticides to control pests and diseases. What’s more, virus-free seedlings compared with non-virus-free seedlings exhibited a positive advantage when cultivation, and they budded earlier than non-virus-free seedlings, had a higher yield, a longer harvest period, and their fruit quality was also better. |
