In the production process, found that there are differences in the number of rib of different individual in the groups of Songliao Black Swine. Combined with the breeding history of Songliao Black Swine, in order to analysis that the difference of rib numbers in the groups of Songliao Black Swine, this research takes the Songliao Black Swine as the experimental object groups and the hybridization pig of Landrace-Yorkshire-Duroc as the experimental control group were collected, the sample of the longissimus muscle tissue including 173 of Songliao Black Swine and 283 of the hybridization pig of Landrace-Yorkshire-Duroc were the test material for extraction of genomic DNA, and according to the sampling sequence recording phenotypic value of rib. NR6A1 and VRTN gene were selected as candidate genes that were significantly associated with vertebral number, using PCR-RFLP and gene typing technology for polymorphisms screening of candidate genes, and further analysis phenotypic value of rib, expecting provide a molecular markers of the close to the ideal and a theoretical basis for further breeding and improve to construct multiple rib strain in groups of Songliao Black Swine. This research results were as follows:NR6A1 gene does not exist gene polymorphism in groups of Landrace-Yorkshire-Duroc, but the presence of polymorphisms in groups of Songliao Black Swine, the presence of A>G mutations in the target fragment 184 bp; the VRTN gene has genetic polymorphism in groups of Landrace-Yorkshire-Duroc and Songliao Black Swine, there exists a 289 bp insertion mutation in the 112 locus of target fragment; Verify that the cutting sites of Nci I enzyme in target fragment of NR6A1 gene which existed within select clear effect in groups of Landrace-Yorkshire-Duroc, but which existed AA, AG and GG three genotypes in groups of Songliao Black Swine, and the locus was a Hardy-Weinberg equilibrium state, which belonged to moderate polymorphism; There wasn,t a significant correlation that 184A>G locus of NR6A1 gene lead to variation on the number of rib on Songliao Black Swine individuals, There was a significant correlation that the insertion mutation at 112 locus of VRTN gene lead to variation on the number of rib on Landrace-Yorkshire-Duroc individuals and Songliao Black Swine individuals, Therefore,the insertion mutation locus may very important to provides a molecular markers of the close to the ideal for further breeding and improve to construct multiple rib strain in groups of Songliao Black Swine.
