Song liao Black pig took a local pig breeds-Northeast Min pig as the first female, landrace as the first Male parent, Duroc pig as the second male parent, cultivated the first black female Porktype pig breed in northern area of China through more than20years after hybridization innovation, cross fixed, breeding improve stage, the pig have the advantages of both the Chinese native pig breeds and the introduction pig breeds. At present, it has not related research of PRLR gene and OPN gene of Songliao Black pig. This experiment takes Songliao Black Pig as the main research object, landrace pig as contrast group,studies polymorphism distribution of two genes and the influence of the reproductive traits of sows, to discuss the feasibility of the use of the two genes as a molecular marker for improving reproductive performance of sow for providing a theoretical basis of Songliao Black pig Breeding to further.This study collected the ear tissue sample of110head of Songliao Black pigs and64head of Landrace pigs,with conventional DNA extraction method for obtaining genomic DNA as template, using6pairs of primers on OPN and PRLR genes amplification,by PCR-SSCP and PCR-RFLP technology studying polymorphism the two genes, using SPSS13.0analyzes genotype frequency and genetic structure of polymorphic loci frequency, and analyzes the relationship different genotypes and reproductive traits of sows. Relevant research results are as follows:1A microsatellite loci was detected in promoter region of OPN gene, and detected six alleles(namely A,B,C,D,E and F) in the microsatellite loci. Five kinds of genotypes(AB, BD, AE, BC, DF) were found in the two study groups. polymorphic loci were not found in the P4and the P5primer region of OPN gene.2P2primer amplification area exist polymorphism site, In the two groups A, B two alleles and AA, AB, BB type three genes are detected, Songliao black pig’s A and B allele frequency0.136and0.418, respectively, but the Landrance pig0.172and0.344, indicating that B alleles is superiority in two groups. The chi-square test shows that two of the group that P2sites genotype distribution has reached the Hardy-Weinberg equilibrium state. Songliao black pig and the Landrance pig’s P2site of polymorphism information content was respectively0.3176and0.4205, belongs to moderate polymorphism. The least square analysis indicated that In the two groups population the number of weaning piglets of AA genotype sows were significantly higher than for AB, BB genotype sows(P<0.05), but no significant difference between the other characters.3In P3site, there detected three genotypes in Songliao black pig population, while only AA, BB two genotypes in the Landrance pig. The A allelics frequency is both higher than B in two groups. The chi-square test shows that Songliao black pig is in a Hardy-Weinberg balance, and the Landrance pig is not in Hardy-Weinberg equilibrium state. The P3polymorphism information content of two groups are bigger than0.5, explaining that it is highly polymorphism in P3sites.Total number born, number born alive, weaning individual weight, number at weaning of the BB genotypes’ Songliao black pig early sow all are significantly higher than those of others(P<0.05); Number at weaning of the AB genotypes Landrance pig is significantly higher than others’(P<0.05).4In P3site, there detected three genotypes in Songliao black pig population, while only AA, BB two genotypes in the Landrance pig.
