By use of the yak as the research materials, two experiments were carried out in the present study. The epithelial cells from yak oviduct were first isolated and cultured using the improved traditional method; and thereafter the effect of Follicle Stimulating Hormone(FSH) on the glycoprotein produced by oviduct epithelial cells were investigated.The oviduct were isolated using different digetive enzymes to dissociae it, and the purified oviduct epithelial cells were collected and indentified The cultured bovine oviduct epithelial cells were treated with different dose of FSH, and the m RNA expression and the secretion site of oviduct-specific glycoprotein were determined.The results showed that:(1) The optimal methold for isolation and culturing of oviduct epithelial cells in yak was that the oviduct should be treated with two-step enzymatic digestion with 1g/L collagenase I-hyaluronidase and 0.25% trypsin. A high purity rates of about 90%would be obtained by use of this culture method.(2) A s-shaped growth curve of the oviduct epithelial cells was found. It can be seen from the curve that the cells grown slowly during the first 3 days, and did exponentially in the fouth to fith day, and again did slowly from the sixth day.(3) Of all the cultured oviduct epithelial cells, they had the ability to express oviduct-specific glycoprotein, mainly by cytoplasm.(4) The m RNA expression quantity of oviduct-specific glycoprotein would raise with the increase of FSH concentration when the FSH concentration maintained within 0.5~5 μg/m L. A highest m RNA expression quantity would be find when the FSH concentration was 5 μg/m L; while the expression quantity would decrease when the FSH concentration was more than 10 μg/m L.The oviduct epithelial cells in yak were cultured in vitro successfully in the present study. We also found that FSH could promote oviduct epithelial cell to secrete oviduct-specific glycoprotein in a dose-dependent ability. This result provide us abasic information for solving low reproductive rate and embryo transfer survival rate,also for further studying on the reproductive biology in yak.
