Prostaglandins are physiologically active substances,which contain various secretions related to the reproduction in active substance.Among PGs,PGE2 and PGF2? had serious function on the area of reproductive physiology where the synthesis,secretion and distribution in the oviduct as well as the possibilities of participating and adjusting the early embryonic development indicated the important factor that are them in oviduct.This study using the oviduct epithelial cells discuss PGE2 and PGF2? on oviductin expression.The details are there:?1?The study of EP2 and FP receptors agonist for influence on the oviductin expression in the oviduct epithelial cells.Using RT-PCR and Westernblot detection of EP2 receptor agonist butaprost and FP receptor agonist fluprostenol had the influence on the oviductin in oviduct epithelial cells.The results showed that butaprost could significantly promote oviductin m RNA experssion in 4 h and 8 h.And also significantly promote the protein expression of oviductin in 4 h,8 h,16 h,and 24 h.After the pre-processing of indomethacin,oviductin expression was decreased.After the addition of fluprostenol,the expression of oviductin was decreased in each time group.But there are not significant difference between indomethacin and fluprostenol group.?2?The study of estrogen and EP2 and FP receptors agonist for influence on the oviductin expression in the oviduct epithelial cells.Using RT-PCR and Westernblot detect butaprost and fluprostenol and estrogen working on oviductin in the oviduct epithelial cells.Expression of oviductin with butaprost and estrogen corporate effect was higher than both of them signally working on.But,the expression of oviductin after adding fluprostenol and estrogen was higher than fluprostenol alone and lower than estrogen alone.?3?The study of PGE2 and PGF2? on the oviductin expression in the oviduct epithelial cells through signal pathway.Using RT-PCR and Westernblot,the use of PKA inhibitor H-89,PKC inhibitor Chelerythrine and ERK inhibitor U0126 treatment of oviduct epithelial cells and detecting of the expression of oviductin.The results of this study showed that 3 ?M H-89 and 3 ?M U0126 could block the role of PGE2 in promoting the expression of oviductin.After adding 5 ?M Chelerythrine chloride and 3 ?M U0126,the expression of oviductin was increased.?4?The study of LPS-PGE2-EP2 receptor and LPS-PGF2?-FP receptor for influence on the oviductin expression in the oviduct epithelial cells.Using RT-PCR and Westernblot,the expression of oviductin is detected by LPS stimulate.The results shown that LPS could stimulate oviductin expression and reach the peak at 16 h.The expression of oviductin with LPS and butaprost effect was higher than both of them signally working on.The effect of LPS and fluprostenol on the expression of oviductin was higher than that of fluprostenol alone,but it was lower than LPS alone.NF-? B inhibitor NAC inhibits the expression of oviductin.Conclusions:?1?PGE2 promotes the expression of oviductin in bovine oviduct epithelial cells by activation of EP2 receptor.PGF2? inhibites the expression of oviductin in bovine oviduct epithelial cells by activation of FP receptor.?2?Estrogen and EP2 receptor had the coroperate function of controlling the expression of oviductin in bovine oviduct epithelial cells.Estrogen and FP receptor had the indirect antagonistic action for the expression of oviductin in bovine oviduct epithelial cells.?3?PGE2 affect the expression of oviductin in bovine oviduct epithelial cell which was related to the EP2-c AMP-PKA signal pathway;PGF2? inhibited the expression of oviductin in bovine oviduct epithelial cell which was related to the FP-Ca2+-PKC signal pathway;PKC,ERK can further activate the PKA pathway.?4?LPS by adding the containing of PGE2 can promote EP2 for raising oviductin in bovine oviduct epithelial cell.LPS by decreasing the containing of PGF2? blocked indirect FP for inhibited oviductin in bovine oviduct epithelial cell.LPS could impact the production of PGE2 through NF-? B which effect on the expression of oviductin in bovine oviduct epithelial cells.
