Soil salinization is one of the world’s most serious environmental and socio-economic problems, beeding salt-tolerant materials and revealing the salt-tolerance mechanism of plants under salt stress environment, what are an important guiding significance for resource development and utilization of saline soil and saline water irrigation of agricultural development. Kenaf(Hibiscus cannabinus L.) has especial characteristics, such as infertile, drought and salinity resistance, easily cultivated, fast growing and high fiber output, what is an potential economic crop of salt tolerance, desalination and high-yield. Jute (Corchorus) is an important industrial raw materials of linen, which has high fiber content and the texture of fiber is soft, which has moisture absorption, breathable, antibacterial characteristics. The purpose of this study is to screen salt tolerance kenaf varieties, establish index evaluation system of kenaf salt tolerance, and through analysis of physiological indicators on different growth stages of kenaf, initially to explore the mechanism of kenaf salinity. The fingerprinting of 34 copies of jute germplasm resource were drawn using SRAP markers, providing a theoretical basis of molecular identification for the jute germplasm resources.1、Identification of salt tolerance of germination of kenaf genetic resources. In order to explore the impact of salinity on germination of kenaf,68 copies of kenaf seed were treated using 0.1%,0.3%,0.5%,0.7%,0.9% and 1.1% NaCl concentration, preliminarily obtained salt concentration of physiological poison, and determined that the 0.3%-0.5% NaCl concentration was suitable for identifying salt tolerance during bud stage. And then,68 copies of kenaf material were divided treated into four categories using 0.4% salt concentration during the buds stage, and 14 copies of salt tolerance material were identified:Jinhang25, Transgenic SA-41, Transgenic SA-42, H316, Bt952-37, Fuhong Hang 5th, Fu hong 992, Transgenic of Yew-4 (green), Fuhong 991-1 (red), Fuhong Hang 992 (prematurity), Fuhong 9th, Fuhong Hang952, Fuhong Hang 992, H293.2、Analyzed growth and physiological indexes of seedling of kenaf under salt stress. Through the study of salt tolerance of germination of kenaf, screening 20 copies of different salt tolerance of kenaf, what were used as the materials of seedling stage. Next, using different NaCl concentration treat those seedlings. The results showed that 0.9%-1.1% NaCl concentration was the suitable concentration to identify the salt tolerance differences of kenaf varieties; Re-determination of the growth and physiological indexes under 1% NaCl concentration, results showed that salt tolerance of germination and seedling of kenaf different and proline been used as the salt injury index. The total average of the membership values of the salt injury rate in the bud and seedling indicators were used to evaluate salt tolerance of kenaf comprehensively, results showed that H316, Transgenic SA-41and Zhonghong kenaf 12th had the best salt tolerance. Significant analysis of each salt tolerance evaluation showed that:the germination potential, germination rate, germination index and survival rate on bud stage, as well as plant height, could be used as comprehensive evaluation index of kenaf salt tolerance, otherwise, fresh weight and main root length could be used as auxiliary indicators.3、To explore the mechanism of kenaf salinity tolerance, Na+, K+ and Na+/K+ in different organs of kenaf which were under the coastal saline and the normal soil conditions were comparatively analyzed, as well as the blade selective transport coefficient (Sk, Na).The results showed the Na+content in different organs under salinity conditions was higher than under normal conditions, and Na+ content was 1.53 times higher than normal conditions in leaf, which was the largest difference; the K+ content decreased in root and stem, but was 0.72 times higher than normal conditions in leaf; the blade selective transport coefficient (Sk,Na) also increased.4、Through screening 35 pairs of SRAP primer combinations,12 high stability, polymorphic SRAP primer combinations were obtained; 34 copies of jute germplasm resource were amplified by 12 pairs of SRAP primers, and a total of 132 polymorphic bands were obtained which polymorphism proportion was 84.85%.; 12 pairs of primers were able to distinguish the 34 jute germplasm resources, the number of polymorphic bands amplified and varieties distinguished were comprehensivly analyzed, and the fingerprintings of 34 copies of jute germplasm resources were established.
