Cucumber green mottle mosaic virus (CGMMV) is a member of the genus Tobamovirus in the family of Tymoviridae, mainly infects Cucurbitaceae crops, causing symptoms such as mottle mosaic, chlorosis and distortion on leaf. Currently, CGMMV has been found in many countries around the world. In China, many provinces have also been reported the occurrence of the CGMMV disease. In order to know whether CGMMV had been spread into Jiangsu Province, an extensive investigation on Cucurbitaceae crops was carried out in 2012. Suspected CGMMV infected symptoms such as chlorosis and mottle mosaic were found on the Cucurbitaceae crops, including watermelon, bottle gourd and pumpkin at Hongze, Yizheng and Dongtai. Later identification results showed that the pathogens of three suspected CGMMV infected samples collected from greenhouse watermelon at Hongze, open field bottle gourd at Yizheng, open field pumpkin at Dongtai were indeed CGMMV.In order to know better about the distribution, occurrence, damage degree and epidemic law of CGMMV in Jiangsu Province, and to provide scientific disease prevention and control, a comprehensive survey was made on Jiangsu Cucurbitaceae crops in 2014. The investigation results showed that CGMMV was widely distributed at more than 20 cities or counties in the south, middle and north area of Jiangsu Province, it caused damage to the growth of the Cucurbitaceae crops including pumpkin, cucumbers and zucchini etc. In brief, CGMMV has become a serious threat for the Cucurbitaceae crops production in Jiangsu Province. We also sequenced the genomes of CGMMV isolate CGMMV-JSHZ12 from Hongze, isolate CGMMV-JSDT12 from Dongtai and isolate CGMMV-HaiN12 just isolated from Hainan and analyzed their possible infection source respectively. Results showed that the coding regions of all three CGMMV isolates were same, but the 5’and 3’NCR (Non-coding region) were slightly different. The genome of CGMMV-JSDT12 was 6423 bp (GenBank accession number KC852072) long and and highly homologous with Hunan isolates. This indicates that CGMMV-JSDT12 may be come from Korea directly or spread into some watermelon seed production areas of China and then spread into Jiangsu. The genome of CGMMV-HZ12 was 6423 bp (GenBank accession number KC852073) long and highly homologous with Henan, Hubei, Shandong, Shanxi, Gansu, Hunan, Xinjiang and Yunnan isolates. This indicates that the CGMMV-HZ12 may come from above areas of China. The genome of CGMMV-HaiN12 was 6425 bp (GenBank accession number KC852074) long and and highly homologous with Hunan isolates. This indicates that the CGMMV-HaiN12 may be come from Korea directly or spread into some watermelon seed production areas of China and then spread into Hainan.
