The objective of this paper was to investigate the effects of three protein feedstuffs (the soybean meal, the rapeseed meal and the cottonseed meal) and four carbohydrate feedstuffs (the paddy rice, the corn, the wheat bran and the rice bran) on quantities of rumen microbe and bacterial community adhensive to solid fractions in Si chuan Xuanhan yellow cattle. Experiment 1 Effect of different protein source feedstuffs on the quantities of rumen microbe adhensive to solid fractions in Xuanhan yellow cattleFour rumen-cannulated Xuanhan yellow cattles with similar healthy and weight (408.56±0.47 kg) were each fed four diets with different protein source feedstuffs over a period in a 4×4 Latin Square design, four periods, each consisting of 7 d adaptation period followed by 3 d collection period,which were basal diet(CN), replaced 15% concentration of basal diet with soybean meal (SBM), rapeseed meal (RSM) and cottonseed meal (CSM). respectively. The PCR technique was introduced to analyze the change of the quantities of ruminal microbe adhensive to solid fractions at 1 h,3 h,5 h,7 h and 9 h after morning feeding. The results showed that:1)the highest value was found in group SBM while the lowest occurred in the group CSM on R.amylophilus(P<0.05):2) the quantities of Prolozoa.R.flavefacien and F.succinogenes in group RSM were significant higher than that of group CSM and group SBM(P<0.05). 4) the quantities of Fungi, B. fibrisolvens, P.ruminicoal and S.Bovis of group CSM were significant higher than that of group RSM and group SBM(P<0.05). Experiment 2 Effect of different carbohydrate feedstuffs on the quantities of rumen microbe adhensive to solid fractions in Xuanhan yellow cattleFive rumen-cannulated Xuanhan yellow cattles with similar healthy (409.28±12.86 kg) were each fed five diets with different carbohydrate feedstuffs over a period in a 5x5 Latin Square design, five periods, each consisting of 7 d adaptation period followed by 3 d collection period, which were basal diet(CN). replaced 20% concentration of basal diet with corn(CD), paddy rice(PR), wheat bran(WB) and rice bran(RB),respectively. The PCR technique was introduced to analyze the change of the population of ruminal microbe adhensive to solid fractions at 1 h,3 h,5 h,7 h and 9 h after morning feeding. The results showed that:1)the highest mean value was found in group CD (P<0.05) on rumen microbe,except the Fungi was significantly lower than group PR and group WB(P<0.05) and R.albus, S.Bovis and S.ruminantium were lower than group PR(P>0.05); the quantities of Protozoa,Methanogens,R. albus, F.succinogenes,R. amylophilus,P. ruminicoal and S.amylolytica in group CD were significant higher than another group; 2) cereal grains energy feedstuffs were significantly higher than bran class feedstuffs on the quantities of rumen microbe (P<0.05). Experiment 3 Effect of different source feedstuffs on reveals diversity in the bacterial community adhensive to solid fractions in Xuanhan yellow cattleThe DNA samples from experiment 1 and 2. PCR-DGGE analysis reveals diversity in the bacterial community adhensive to solid fractions in Xuanhan yellow cattle.DGGE profiles showed:1)in all feedstuffs, the highest bacterial diversity was found in group RSM.2) Analysis from sequences showed that the common bands belonging to lactobacillus, Uncultured Bacteroidetes bacterium, Uncultured Bacilli bacterium and Uncultured rumen bacterium. The Specific bands that CD and WB without belonging to Streptococcus and Uncultured Firmicutes bacterium.In conclusion, different protein feedstuffs and carbohydrate feedstuffs significantly affected the quantities of rumen microbe and bacterial community. Cottonseed meal significantly promote ruminal proteobacteria adhensive to solid fractions, rapeseed meal significantly promote ruminal cellulosis microbes adhensive to solid fractions; corn have a better promoting effect on rumen microbe. Protein feedstuffs were more affected bacterial community adhensive to solid fractions than carbohydrate feedstuffs; Analysis from sequences showed that the common bands belonging to lactobacillus, Uncultured Bacteroidetes bacterium, Uncultured Bacilli bacterium and Uncultured rumen bacterium.
