| Chicken in China are abundant in genetic resources, Chinese local chicken of obvious characteristics have been formed after several thousands year development. However, we are still facing the problem of low integrated laying performance and the loss of genetic resources. In order to protect and utilize layer genetic resources well, our study is to use the green shell layer and Barred Rock layer as the research object. The laying performance of 200 green shell layers and 200 Barred Rock layer were measured. We recorded the time of laying first egg and the egg production of 300 days age. According to the SNP detection, we analyzed the correlation of ER-a, the polymorphism of VIPR-1 and laying performance. The results are as follow:1. Laying performance comparison and measurement between green shell layer and Barred Rock layer.The age and the weight of first egg of green shell layer is 162.60±11.38d and 33.72 ±7.31g. The egg production, the egg weight and the egg yolk percentage within 300d of green shell layer is 83.77±20.44,51.03±4.57g and 30.65±1.77%.The color and the weight of egg shell is 42.71±5.22 and 5.61±0.70g. The height,weight and proportion of protein is 3.85±0.86mm,29.03±2.15g and 58.19±2.44%.The age and the weight of first egg of Barred Rock layer is 154.50±8.42d and 44.27±6.41g. The egg production, the egg weight and the egg yolk percentage within 300d of Barred Rock layer is 116.37±13.75,61.30±4.15g and 25.30±2.70%.The color and the weight of egg shell is17.07±3.40 and 6.40±0.58g. The height,weight and proportion of protein is 7.13±1.20mm,39.07±3.46g and 63.44±3.27%.According to the result, the green shell layer is significantly higher in egg yolk percentage and darker in egg color than Barred Rock layer. But the height,weight and proportion of protein are significantly lower than Barred Rock layer. The laying time of green shell layer is 8d later and the weight of first egg is 10.55g lighter compared to Barred Rock layer. Eggs within 300d, the egg weight of Barred Rock layer is lOg lighter and the egg production is 31 fewer than the green shell layer. All these results reached very significant difference level. These indicators provide the basis for the next step of breeding work.2.Correlative analysis of laying performance and gene SNP polymorphism of ER-a gene.In the region of the ER-α gene intron 1, found two SNP locis,the loci were A27327G and C27347T, and formed four haplotypes.And compare the two SNP and laying performance relationship,in SNP1 loci,allele AG is the dominant type in age of first egg(P<0.05), and allele AG, GG are the dominant type in quantity and weight of eggs within 300d(P<0.01);On SNP2, allele TC is the dominant type in quantity of eggs within 300d(P<0.05).So, the SNP polymorphisms in ER-a gene intron 1 is significant effect on laying performance,based on ER-α polymorphism and the chicken laying performance relationship studies by the predecessors.it could offer theory evidence for ER-a as molecular marker assisted selection.3.Correlative analysis of laying performance and gene SNP polymorphism of VIPR-1 geneIn the region of the VIPR-1 gene intron 1,found three SNP locis,the loci were T38181G〠A234G and A345G, and formed six haplotypes.And compare the three SNP and laying performance relationship,in SNP1 loci,allele GT is the dominant type in age of first egg(P<0.05y),and allele GT, GG are the dominant type in quantity and weight of eggs within 300d(P<0.05);On SNP2, allele AA,AG are the dominant type in in age of first egg and quantity of eggs within 300d(P<0.05,allele AA is the dominant type in weight of eggs within 300d(P<0.05),and the result of SNP3 loci is consistent with SNP2 loci.So,the SNP polymorphisms in the region of the VIPR-1 gene intron 1 is significant effect on laying performance,based on the VIPR-1 gene relationship studies by the predecessors.it could offer theory evidence for VIPR-1 gene as molecular marker assisted selection. |
