Tartary buckwheat is an ancient annual dicotyledonous herb, which is widely distributed around the world. It has strong adaptability and environmental tolerance characteristics. It originated in China and it has excellent nutritional value. It has presumed to be involved in many of the health benefits and special medicinal properties. It is also the raw material for many green and important functional foods.In our study, we used Yuanzi, No.l chuanqiao and Jiujiang tartary buckwheat as our experimental materials. Finally, we established an efficient hairy roots culture system of tartary buckwheat which induced by Agrobacterium rhizogenes using the cotyledon and hypocotyl as explants. Meanwhile, a number of factorshave been discussed.We found A. rhizogenes A4 has the maximal induction efficiency of hairy roots in three different A.rhizogenes strains we used. And Yuanzi tartary buckwheat has the maximal induction efficiency of hairy roots in three different experimental materials. After two days pre-incubation, the highest induction rate is 83.6% when we used A.rhizogenes to infect the Yuanzi tartary buckwheat hypocotyl.Moreover, we used Yuanzi and Jiujiang tartary buckwheat as experimental materials, and an efficient tartary buckwheat regeneration system via somatic embryogenesis was established.Finaliy, the regeneration frequency of Yuanzi tartary buckwheat was best and the regeneration frequency of hypocotyl was the highest in three different tartary buckwheat species. We used 2.0 mg/L 2,4-D and 1.0 mg/L KT on MS medium in callus induction phase, the best callus induction frequency is 98.94%. And then we used 1.0 mg/L KT and 2.0 mg/L 6-BA on MS medium in embryoid buds induction phase, the best buds differentiation rate is 55.53%. After that we used 1.0 mg/L IBA on 1/2MS for regeneration rooting, the best rooting rate is 86%. In this case, wecanachieve a better tartary buckwheat regeneration system.We also used apical meristem of tartary buckwheat as explantfor plant regeneration via organogenic pathway simultaneously. The result shows that:If adding 2.0 mg/L 6-BA and 0.5 mg/L TDZ in the MS culture medium, we can get the best regeneration frequency via multiple shoot induction. The best buds differentiation rate is 69.05% and the largest number of budding is 18.
