This paper studied industrial production condition of Bacillus coagulans BCRC11592 preserved by our laboratory,which has been shown to improve the growth performance of piglets. It was proved that it could tolerate heat,acid,bile salt. Optimize the medium component,inoculum age and loaded liquid of the bacteria. Based on a more systematic media and fermentation conditions, the influence of stirring speed,throughput and constant p H for cell growth and spore was analysed by the production test. Concentrate and adsorb the fermented liquid to get condensate bacillus viable pharmaceutics. Then simulating pelleting temperature, digestion test in vitro,drug sensitive test and storage test could be used to preliminarily evaluate the production. All these above meant to provide the theoretical basis and data bases for the development and application of probiotics.Experiment 1: the optimization of fermentation conditions and cultural media for Bacillus coagulans BCRC11592After activating Bacillus coagulans BCRC11592, the single factors and orthogonal test were carried out to optimize the industrial medium of Bacillus coagulant. The single factors were inoculum age, carbon source, nitrogen source,and inorganic salt. The orthogonal test of four factors at three different levels were used. At last The best loaded linquid have been found. The optimal medium were as follow:corn meal 5g/L、wheat bran 10g/L、NH4Cl 7.5g/L、Soya bean meal 7.5g/L、 Na Cl5g/L、Mn SO4·H2O 0.3g/L、K2HPO4·3H2O 1g/L. Because of the test, the optimal loaded liquid(35ml)was chosen. The viale count of Bacillus coagulant was 2.20×109 and the number of endospore was 2.02×109 by flask fermentation.Experiment 2: The preparation technology of condensate bacillus viable pharmaceuticsSteaming and filtrating substrate raw materials to prepare the fluid nutrient inthe fermentor. The media was disinfected, then made cool, inoculated 1% seed broth. The proper operation condition was 200r/min of RPM, 350L/h of throughput and don’t balance p H. Concentration process by micro-filtration was 0.14~0.16 MPa of operation pressur, 37℃~40℃ of temperature and 40 min of time.The broth concentrated by 2.86 times from micro—filtration. Mineral adsorbent adsorbed the concentrated solution by 1:1.Experiment 3: The product evaluation of condensate bacillus viable pharmaceuticsThe results of tests showed that the survival rate of the strain were respectively 71% after processing the pelleting temperature of 85℃for 10 min; the survival rate was 86.8% after processing the simulated gastric fluid; the survival rate could reach to 69% after experiencing simulated intestinal fluid; Drug sensitivity tests showed that the product could be used with amoxicillin; and the survival rate were more than 50% after processing the temperature of 25℃ and35℃ for 30 days, beside the quantity of the strain was 108 orders of magnitude after processing the temperature of 25℃ and 35℃ for 60 days.
