Tilmicosin is tylosin by ortho-oxybenzoic acid of derivation, it is macrolide livestock antibiotics. It has broad antibacterial effects and high antibacterial effects or low toxicity. In clinical veterinary practice tilmicosin is mainly used to treat cattle, goats, pigs, chickens and other animals diseases, especially the poultry respiratory diseases. However, because of not readily soluble in water, the efficiency of tilmicosin by oral administration and injection can be affected. In order to widen the application range of tilmicosin in veterinary clinic. The aim of this study was to investigate 10% pharmacokinetics of phosphate tilmicosin in broiler chickens under the establishment of the detection method of phosphate tilmicosin soluble powder.The detection method of High performance liquid chromatography (HPLC) was used to separate and purify the phosphate tilmicosin in the serum of chicken. Under the testing conditions of mobile phase of acetonitrile:tetrahydrofuran:dibutyl amine phosphate buffer:water=10:5:5:80, C18 column (4.6×50 mm,5 μm), the flow rate 1.0 mL/min, the column temperature 30℃, detection wavelength at 280nm, the results showed the good linear relationship between the peak area and the corresponding concentration, the average recovery is above 76.9%. The detection methods of the phosphate tilmicosin in broiler chickens established by HPLC in this stuy is simple, precise, fast and easy to operate.The detection methods of pharmacokinetics of the phosphate tilmicosin in chickens were established in this study by high performance liquid chromatograph (HPLC). It has with 10% phosphate tilmicosin soluble powder high dose (16 mg/kg)、medium dose (8 mg/kg)、low dose (4 mg/kg) and 10% control tilmicosin soluble powder (8mg/kg) by broiler chicken oral medicine. The results showed that the average means of area under plasma concentration-time curve (AUCo~∞) for high dose (16mg/kg), medium dose (8 mg/kg), low dose (4 mg/kg) of phosphate tilmicosin and for 10% control Tilmicosin in broiler chickens were 5.6163±0.9584,2.6082±0.2665,1.4716±0.0869,2.3314±0.1956,the results showed that proportional to the amount of given medicines,which was almost proportional to the dose and indicating that the pharmacokinetics of Tilmicosin phosphate soluble powder in broiler chickens is the first-order Kinetic process. After taking 10% phosphate tilmicosin soluble powder medium dose (8 mg/Kg) and 10% control tilmicosin (8 mg/Kg), Cmax respectively was 0.2658±0.0205μg/mL and 0.2336±0.0337μg/mL, in a range of bioequivalence (0.80-1.25). The results also showed that tilmicosin phosphate was absorbed quickly, distributed widely into the tissues of broilers and eliminated slowly. According to the important evaluation index of bioequivalence, phosphate tilmicosin soluble powder and control tilmicosin soluble powder can be used interchangeably in veterinary clinics. In addition, intravenous injection of 10% phosphate tilmicosin (5 mg/Kg), all tested broiler chickens were died immediately, it is recommend that Tilmicosin phosphate can not be used to broiler chickens by intravenous injection.
