This paper mainly studied that the cultivation of Grifola frondosa by JUNCAO technology, compared and analyzed the basic nutrient content of Grifola frondosa fruit bodies by different culture medium; Also studied on the extraction and purification of the polysaccharide from Grifola frondosa by JUNCAO,at the same time the polysaccha-ride from Grifola frondosa cultivation with sawdust was compared with,the main findings are as follows:1. The optimum JUNOAO medium of Grifola frondosa was carried out as follows by the screening test, the optimum formula of cultivation composts was Miscanthus floridulu:50%, Dicranopteris dichotoma:28%, Bran:5%, Corn flour:15%,Gypsum:2%, Moisture content: 60%.2. This paper compared and analyzed the basic nutrient content of Grifola frondosa fruiting bodies by different culture medium.The main research results were as follows: the crude protein, crude fat, ash and amino acid content of Grifola frondosa fruiting bodies of the best JUNCAO formula cultivation were higher than the Grifola frondosa by sawdust cultivation, moreover the total sugar content was slightly lower. The heavy metal content of Grifola frondosa cultivated by JUNCAO was in line with national health standards.3. It was determined for the the extracting process of polysaccharide of Grifola frondosa by orthogonal experiment. The results showed that:when polysaccharide were extracted by hot-water extracting method, The impact of every factors on the rate for extraction of the crude polysaccharides was different, the order of effect was solid-liquid ratio,extraction temperature, extraction times and extraction time. The optimum extracting conditions were ratio of material to water 1:40, temperature of 100℃,time of 2 hours and extract 2 times.4. DEAE-Sephadex A-25 chromatography was used for isolating and purifing polysaccharide of Grifola frondosa cultivated by JUNCAO and cultivated by sawdust.The results indicated that:each polysaccharide had both two components,they were C-GLP1 C-GLP2 and S-GLP1, S-GLP2.DEAE-Sephadex G-200 chromatography was used for purity identification of C-GLP1 and S-GLP.The results showed that they were both single component.
