Grifola frondosa, is a rare edible and medicinal fungi which can biosynthesize polysaccharide of bioactivity and less poisonous side-effect on our health during its liquid culture. The liquid culture of Grifola frondosa is a new approach to develop new health food for epiphyte polysaccharide and has prominent economic value. This study explored the composing of culture medium and the condition of liquid fermentation on biosynthesizing polysaccharide of Grifola frondosa. And studied the condition of extracting polysaccharide and purification by orthogonal experiments. These offered theories to develop new health food of Grifola frondosa. Also established bases for making good use of polysaccharide resources from Grifola frondosa mycelium and from edible fungus in biology reactor.In order to determine the content of polysaccharide in Grifola frondosa mycelium, the author studied the anthraceneketone colorimetry. As anthraceneketone reagent can react with furfural and its ramification which was created from the reaction of polysaccharide and vitriol . The reaction can create cyan compound which had the most absorption on 610nm, and the absorption cost had linearity relationship with the content of polysaccharide.The standard curve was set up, and the regressed equation was: y = 0.112x + 0.0154,R2 = 0.9982(x- absorbency on 610nm, y-the consistence of glucose (μg/mL )).By the single factor experiments and orthogonal experiments, the optimum culture medium was established. The influence of carbon resource and nitrogen resource in culture medium on polysaccharide-accumulating fermentation were studied respectively. On the basis of the single factor experiments, orthogonal experiments were performed on carbon resource (potato/glucose),nitrogen resource (bran),abio-salt (KH2PO4 and MgSO4) and growth-accelerant (yeast cream) with dry weight of mycelium and the content of polysaccharide as criterion.
