The Bacillus amyloliquefaciens species is a kind of bacteria, high affinity with Bacillus subtilis and distributed widely in nature. It is able to exert inhibiting effect on various pathogenic fungal and bacteria. Meanwhile, Bacillus amyloliquefaciens can produce rich metabolites in its growth process. Therefore, Bacillus amyloliquefaciens has favorable application prospect in biological prevention and control, and with the development of molecular techniques, gene function of Bacillus amyloliquefaciens will be better understood. The research shows that even the same Bacillus species are significantly different from the mechanisms of antimicrobial action. By comparing the morphology, biological activity, and biofilm formation ability of eight B.species, four obviously different B.species were selected. Then Solexa Genome Analyzer technology was further used to conduct the whole genome sequencing of the four selected species so as to compare genomic differences and similarities; by using comparative genomics methods, the differences in genome and evolutionary relationship of main antibacterial material were analyzed. The main results are as follows:(1) Eight isolates of Bacillus species were belong to Bacillus amyloliquefaciens, through alignment analysis the homologous sequence of 16s rRNA sequences and gyrB gene sequences.(2) Observing and measuring cell size, and spore forming conditions of such 8 B. species, with statistical analysis, results manifested that 8 B. species were not only different in morphology, but also in size. Gap between the maximum JT81 and the minimum JT261, T457 is 1.3 μm.(3) Research activity differences protease, chitinase and siderophore produced by 8 B. species as well as their antagonistic ability. The results showed that 8 B. species were not producing chitinase, JT261 and T455 exhibited stronger abilities of protease, JT84 manifested better ability of siderophores; JT81 and T429 had strong ability in biofilm formation; JT81 and the extracellular material of T455 had higher antimicrobial activity.(4) Through the morphological and biological activities of 8 B.species, four Bacillus amyloliquefaciens JT57, JT301, T429, and T455 were selected.(5) The selected species were whole genome sequenced by using Solexa Genome Analyzer technology. The sequencing results then will be multi k-mer matched by using the software of SOAPdenovo. And then specific primers were designed for PCR to fill the gap, which were used to draw fine genome maps of such four species.(6) Understand the differences in gene sequence and evolutionary relationship of main antibacterial of such four B. amyloliquefaciens, through using method of comparative genomics analysis.
