Salmonellosis is one of bacterial disease which is a serious threat to the modern poultry industries. The disease can not only cause mass mortality of chicks, but also slow the growth of chicken and decrease production, resulting in significant economic loss to aquaculture. Vaccination and antibiotics and other drugs are the mainly preventive measure, but they will cause resistance to pathogens and antibiotic residues in poultry products and other issues. It has been found that the incidence of salmonellosis associated with genetic factors of the individual. Studies have shown that polymorphisms NRAMP1, TLR4 and MHC, etc. are associated with the anti-Salmonella pullorum. MyD88, which plays an important role in the immune response, innates immune pathway gene as an important adapter protein. But the relationship between the MyD88 gene polymorphism and pullorum disease has not been reported.In order to explore the relationship between the MyD88 gene polymorphism and pullorum disease. This study firstly analyzes the purifying effect on the base generation, the first generation, thye second generations of lines SD02 and SD03 in Erlang Mountainous chicken, and analyzes the purifying effect on descendant growth and production performance combined with production data. Secondly, based on case-control experiment design,128 positive chicken blood samples were collectted,156 negative blood samples were collectted. The genomic DNA of all the samples were extracted, and then all the genomic was directly sequenced to detect single nucleotide polymorphisms of the CDS area in MyD88 gene, finally, the genotype frequencies, haplotype group build and haplotype were analyzed by using SPASS 21.0 software and Hploview 4.2 software. The results show that:After sequencing, eight SNPs, of which seven were known, were found in the 501 bp of fifth exon fragments, the new one was named SNP8 (G1116T). Only the SNP1 (c.934C>T) allele frequencies in sick-the control group was significant difference (P <0.05), SNP1 (c.934C> T), SNP3 (c.1001G> T) two mutations locus genotype frequencies in the case-the differences in the control group was significantly different (P<0.05), c.934 C> T, c.1001 G> T, c.1010 T> C locus haplotypes composed in pullorum case-control group significantly different (P<0.01). the haplotype of c.1020 G> A, c.1037 G> C, c.1096 A> G, c.1116 G> T locus had no significant difference with anti pullorum trait (P> 0.05).All the results show that:Single nucleotide polymorphisms of chicken MyD88 gene was significantly associated with anti-Salmonella pullorum traits, it can be used as an anti-pullorum trait marker. These results provided a theoretical basis for future research on quality chicken breeding marker assisted selection.
