Strong broodiness and poor laying performance are two important factors to response for inhibiting the development of goose industry. As an important nerve endocrine gland in waterfowl’s brainthe pineal gland controls the process of egg laying and Broodiness directly or indirectly by turning the environment light stimulation into the neuroendocrine signal. In order to explore the mechanism of goose pineal gland’s differentially expression pattern,the pineal gland of goose,during laying and broodiness period, is employed as our study’s experiment materials.Meanwhile, digital gene expression (DGE) Profiling is used to screen these differentially expressed gene.Then analyzing their GO/Pathways enrichment.The main findings are as follows:(1)We sequenced 1.85 G and 1.51 G raw data in the goose library of egg laying and broodiness respectively, and obtained 1.82 G and 1.48 G Clean tags after a series of quality filtering.(2)Using the software of RSEM, we analyzed the gene mapping of two groups respectively, Results showed that the positioning percentage of two groups were higher than 90%(The egg production groupsis 89.49%,the nest group is 89.84%).(3)A total of 99 differentially expressed genes were screened by DGE,and20 genes got positively regulated,79 genes got negatively regulated between them.(4) 95 metabolic pathways are enriched as significantly differectially expressed between two groups.And the main pathways are Tryptophan metabolism, Phototransduction, Serotonergic synapse, Glycerolipid metabolismand Folate biosynthesis. All of these indicatethat the pineal has an important role in controlling the capability of egg laying and broodiness in goose.(5) Differentially expressed genes are mainly enriched in sensory perception of light stimulation,thepineal gland of sensory perception is a key factor in regulating the goose nest process.(6)Obtained network diagram of differentially expressed genes in the pineal gland,thedifferentially expressed genes involved in Metabolism of amioa acid, Metabolism of nucleotide, Metabolism of vitamin, Energy Metabolism, Lipid Metabolism, Glycan Biosynthesis and Metabolism, Carbohydrates Metabolism and so on. Significantly enriched in the Phototransduction, plays an important role in theretinol metabolism.
