| As one of the three diseases of rice to rice blast, which is seriously affecting the important diseases in rice production.In the present study showed that the plant miRNA can regulate the growth and development of plants, biological and abiotic stress, and resistance reaction.But for the miRNA regulation rice and rice blast fungus rarely research report, this laboratory identified the two is the blast resistance of miRNAs:miRNA398 and miRNA 160, and they may be through inhibiting the transcription of target genes to control rice blast resistance.To study miRNA 160 target gene LOC_Os06g47150 and miRNA398 target gene LOC_Os04g48410 biology function, verify the relationship between target gene LOC_Os06g47150 and miRNA160, target gene LOC_Os04g48410 and miRNA398 relations, and further explains miRNA398 and miRNA 160 preparation for the study of the control network.This topic by building miRNA 160 target gene LOC_Os06g47150 and miRNA398 target gene LOC_Os04g48410 expression vector, and the mutation expression vector, and RNAi vector, and genetic transformation in rice to obtain the corresponding materials.In this study, the first is to build a miRNA160 target gene LOC_Os06g47150 and miRNA398 target gene LOC_Os04g48410 expression vector, by injecting the observation of the two genes transient expression in N.benthamiana, found the target genes LOC_Os04g48410 expressed mainly on the cell membrane, and target genes LOC_Os06g47150 mainly expressed in the cell nucleus. Second is to build miRNA160 target gene LOC_Os06g47150 and miRNA398 target gene LOC_Os04g48410 mutation expression vector, and through the method of agrobacterium mediated transformation of rice, after hygromycin identification and PCR identification, LOC_Os06g47150mutant got 4 strains of transgenic rice plants, LOC_Os04g48410 mutant 24 strains of transgenic rice plants. Finally is the target gene miRNA160 LOC_Os06g47150 and miRNA398 target genes LOC_Os04g48410 RNAi carrier construction. |
