First the study is surveying several persimmon varieties of Henan province, then baseing information archives by phenotypic analysis to 8 resources of persimmon varieties of Taihang mountain area in Huixian, finally analysing relationship and geneticing differences about 65 persimmon germplasm of Henan in the national germplasm resources garden of persimmon and 8 persimmon germplasm resources of Taihang mountains in Huixian using SSR(Server Security Reinforcement) molecular markers technique. Through analysis of phenotypes and markers of genomic DNA to the persimmon varieties of Taihang mountain area in Huixian, explore the relationship and genetic diversity between persimmon varieties in Henan province. The main results are as follows:1. Through the literature review and field surveys, collecting informations and taking photoes about 82(6 persimmon resources in Anyang area, 18 persimmon resources in Xinxiang,1 Shi resources in Jiaozuo, 7 persimmon resource in Zhengzhou, 1 persimmon resources in Sanmenxia area, 19 persimmon resources in Luoyang, 7 persimmon resources in Nanyang, 23 persimmon resources in Pingdingshan) persimmon varieties of several persimmon production areas in Henan province, it is found that the number of persimmon varieties is so many, and varies in shape, size, quality and maturity;2. Through the investigation to 8 major resources of persimmon varieties of Taihang mountain areas in Huixian, establish archives informations under the standards of descriptors and data of persimmon germplasm resource, such as their phenology, biological characteristics, appearance, food features;3. Establish the best SSR-PCR reaction system in persimmon varietiesis. PCR amplification is performed on the genome PCR. The total volume of PCR reaction system is 20 μL, contains 2.0 μL 10×PCR buffer, 50 ng template DNA, 0.16 mmol/L d NTPs, 2.0 mmol/L Mg2+, 1.5 U Taq DNA polymerase, 0.4 μmol/L upstream and downstream primer, residual volume filled with sterile double steaming water. PCR reaction procedure: 94 ℃ predegenerated 5 min, 94 ℃ denaturation 45 s, annealing 45 s, 72 ℃ extension 2 min, 35 cycles, 72 ℃ final extension after 7 min and saved at 4 ℃;4. 10 polymorphic primers were selected from 30 pairs primers, which is clear and high polymorphism. Get 68 polymorphism SSR-PCR amplification were obtained using the 10 primer and the genome of 73 persimmon resources, the percentage of polymorphism is 85.35%;5. To calculate genetic similarity and genetic distance of the inter-group using POPGENE 1.32 software, found that the similarity of 73 persimmon varieties resources tested is between 0.3382~0.9904, and the range is large. So it explains persimmon varieties is wide in source and rich in genetic diversity;6. The Treeplot tree obtained by UPGMA cluster analysis using NTSYS PC-version 2.10 E software, and found that 73 persimmon varieties resources can be classified into 2 main SSR group on 0.53 similar factor levels. First group has 59 Diospyros species resources including ‘Hui shi of chengou in Xingyang’, ‘Laomendun’, ‘Guilianqing of Luoyang ’, ‘Laochutou’,‘Tiepi shi’and so on; Second group has 14 persimmon varieties resources including ‘Guilianqing of Boai’, ‘Niangun shi’, ‘Huoguan of Xingyang’, ‘Tiansheng-01 of Yanshi’, ‘Shuiniuxin’and so on. On 0.56 genetic similar factor level, first group can be divided into 2 subgroups, the one subgroup have 57 persimmon varieties including ‘Xiaodishi’, ‘Qiyuezao’, ‘Jingmian shi’, ‘Dabiegai shi’, ‘Changshengtian shi’, ‘Mopan shi’ and so on; another subgroup have two persimmon varieties including ‘Mimiguan of Luoyang’and ‘Yuanguanhong of Zhengzhou’in Henan.
