AFLP (amplified fragment length polymorphism) is a new DNA molecular marker technique with high efficiency, promptitude, stability and accuracy. It has been widely used in classification, variety identification , genetic mapping, breeding, location of target gene in animals and plants.Persimmon(Diospuros kakai L) originated in China and has been cultivated for a long time in China, Korea and Japan. It has rich germplasm and widely distributed in China. Understanding the relationship of different persimmon cultivars and their genetic diversity will be benefit to the utilization of the germplasm and improve the breeding work. In this study, AFLP analysis system fit for persimmon was constructed and the fingerprinting of some excellent cultivars were established. The genetic diversity and genetic relationship of persimmon cultivars had also been analyzed, he main results were as follows:l.The modified CTAB procedure: l00mmol/L Tris-HCI, (pH8.O), 20mmol/L EDTA, 1.4mol/L NaCl, 2% CTAB, 1%PVP40;l00mmol/L β-MMercaptoethanol, 10mmol/INa2S2O5 is suitable for the genomic DNA isolation of persimmon for AFLP analysis.2.A suitable system for AFLP analysis of persimmon was put forward. First, 450ng of genomic DNA was digested with 3 Units of EcoR I and Mse I for 3 hours, then the DNA fragments linked to EcoR I and Mse I adapters. After preselective and selective amplified, the PCR reaction products were analyzed on 6% denaturing polyacrylamide gels and then stained by silver.3.Six primer combinations selected from 64 primer combinations were used for AFLP analysis, based upon the number of scorable strong bands and the level of polymorphisms. 328 bands were scored and 322 bands wre polymorphic. The fingerprinting of 56 persimmon cultivars and 4 varieties of D. lotus L. were established basing on these bands.4.60 persimmon materials used in this study were divided into 2 groups by Ward's method. Their genetic diversity was discussed.
