Racoon dogs with high economic value, its main products fur, it is one of three international fur.Racoon dogs skin belongs to fine leather, with soft, lightweight, durable, beautiful thermal insulation and other features, is a high-quality raw materials for manufacturing high-grade fur coat, fur mattress, cap, collar and other. Its fur quality directly affects the economic benefits generated by the racoon dog, but the emergence of rigid disease seriously affecting the quality of fur, bringing huge economic loss provision racoon dog industry. At present, there are few reports on the causes for rigid disease in racoon dogs, remains to be studied the experimental personnel. This study mainly from two angles of genetic factors and detection of intestinal bacteria population changes of the racoon dog rigid disease causes are explored. This experiment mainly by the tail analysis with 180 racoon dog individual samples for analysis and research, in order to find and rigid disease in racoon dogs associated molecular markers, and found that healthy racoon dogs and stiff racoon dogs in the genetic difference. The extraction of the 30 healthy racoon dogs and 30 stiff racoon dogs’ intestinal bacteria group and by real-time fluorescence quantitative PCR to detect the healthy racoon dogs and stiff racoon dogs in lactic acid bacteria, bifidobacteria, quasi, Escherichia coli, Enterococcus faecalis, Clostridium, Veillonella quantity difference. The main research contents of the experiment are as follows:(1)By tail analysis method and 120 random primers to detect healthy racoon dogs and stiff racoon dogs and find differential markers.(2) To find the specific fragments were cloned and sequenced.(3)By finding specific primers Harbin Huaqiang and Daxinganling breeding base racoon dogs detection(4) By real-time fluorescence quantitative PCR to detect the healthy racoon dogs and stiff racoon dogs in lactic acid bacteria, bifidobacteria, quasi, Escherichia coli, Enterococcus faecalis, Clostridium, Veillonella quantity difference.The main results of the experimental study are as follows:(1) The healthy racoon dogs and stiff racoon dogs exist differences of genetic marker.Preliminary judgment HA12-500, HB19-700 labeled sequence specific sequence of healthy racoon dogs and that is stiff racoon dog deleted sequences.DA19-700, DC7-700 and DF20-1000 to determine the initial mark sequence specific sequence of stiff racoon dogs.(2) Stiff racoon dogs gut flora of Lactobacillus and Bifidobacterium number was significantly lower than that of healthy racoon dogs, Escherichia coli, Enterococcus faecalis, Clostridium number was significantly higher than that of healthy racoon dogs, suggesting that gut microflora dysregulation may is rigid disease in racoon dogs one of the triggering factors.
