Wintersweet(Chimonanthus praecox L.) is popular in China as a traditional plant. It is widely used in the cut flower industry and gardening. The fragrance of flower is the most important feature for wintersweet, for taking the warmth and hopes to people in cold winter. ODORANT1 as the first transcription factor in the biosynthesis of fragrance is the regulation of volatile benzenoids in petunia flowers. CpODORANT1 and CpODORANT2 are transferred into Arabidopsis and tobacco to investigate it’s function.1) Various expression differences of CpODORANT1 and CpODORANT2 are showed in different floral stage of Chimonanthus praecox. The expression of CpODORANT1 in H64 is significant higher than SW001. In other development stages,SW001 is higher than H64. However, the expression of CpODORANT2 shows an opposite pattern. The expression of CpODORANT2 is higher in SW001 compared with H64, but is lower in other stages.The expression of CpEPSPS of different floral periods in genotype H64 are higher than in genotype SW001.2) The expressions of NtEPSPS1 and NtEPSPS2 show no difference in PBI121-CpODORANT2 and PBI121-CpODORANT1 transgenic tobacco plants. Their expressions both rise in stem and leaf in PBI121-CpODORANT2 transgenic tobacco plants; however, in PBI121-CpODORANT1 transgenic tobacco plants, the expressions of NtEPSPS1 and NtEPSPS2 in stem both increase, and in leaf NtEPSPS1 shows no difference, NtEPSPS2 decrease contrarily.3)Compared to wildtype Arabidopsis,The expression of AtEPSPS are both higher in stem, buds and early blossoms in transgenic plants. But it’s expression in full blossoms is lower in transgenic plants.4) The content of Dimethyl benzaldehyde in PBI121-CpODORANT2 and PBI121-CpODORANT1 transgenic tobacco plants is increase. And they contain Cyclotetradecatriene-1,3-diol\Alloaromadendrene\ Germacrene uniquely.5)CpEPSPS are cloned from Chimonanthus praecox. CpEPSPS encodes an open reading frame(ORF) of 1557 bp corresponding to a protein of 518 amino acid and 828 bp promoter fragment before the initial codon is attained.
