The new non-1BL/1RS translocation male sterile line msBi-V9125 carrying Ae.bicornis cytoplasm( short for ms Bi-V9125),which completely to sterile, without anther exposed during flowering stage and haploid produced, and with strong recoverability, showed a new way to study on and make use for heterosis of wheat. To analysis steril mechanism of male sterile line, the F1 of sterile line and maintainer and the F1 of sterile line and restorer line T63 were used in this study. Some preliminary study was done by paraffin section, Label-free and qRT-PCR. The conclusions were below:From the results of paraffin section, the tapetum degradation in male sterile line was earlier than that in fertile. The complete disintegration of tapetum in male sterile line was happened at late unicellular stage, while it in fertile line wasn’t happened still double nucleus stage. Howerer, the differences were obvious between microspores in male sterile line and fertile line. So it was inferred that earlier degradation of tapetum was some influence on the development of microspores, and late unicellular to trinucleate stage was the key stage in the development of microspores.Through Label-free, 2142 proteins were dentified. Amog them, there were 50 proteins only in sterile line and 285 proteins was only in fertile line. All proteins were blasted in mitochondrial protein libraries and 84 mitochondrial proteins was got. There were 2 proteins only in sterile line and 3 proteins only in fertile line. At the same time, all proteins were analysised by subcellular localization. There were 100 proteins localized to mitochondria. The results by Go analysis showed that differential proteins related to many cellular processes,mainly about metabolic process and biological regulation, existed in all cell components,and their molecular function was mainly about regulation and catalyze activity.The results of qRT-PCR shwed that the genes which assigned nad2, COX1, SUV3,ATP6 protein were different significantly between sterile line and fertile line. In msBi-V9125,the expressin of nad2 was rised gradually from tetrad stage to late unicellular stage and droped gradually from late unicellular stage to trinucleate stage. But it was on the contrary in sterile line. The expression of COX1 in sterile line kept low from tetradstage to trinucleate stage. But it was much higher than every stage at tetrad stage in fertile line.The expression of SUV3 was high at tetrad stage, binucleus stage and trinucleate stage, while it was low at uninucleate stage in fertile line. And it kept low in sterile line. The expression of ATP6 in sterile line kept low from tetrad stage to late unicellular stage, but it increased significantly at binucleus stage, and it was a little lower at trinucleate stage than binucleus stage.
