Agrilus zanthoxylumi(Li)(Coleoptera: Bupresdae) is the main stem borer of Zanthoxylum bungeanum Maxim in the pepper producing areas in the north of China. At present, we find Agrilus zanthoxylumi only damaged Zanthoxylum bungeanum, and has caused gummosis,rot even death of the Z. bungeanum,which bring about the decline of production and quality in Z. bungeanum. In the research, the volatile components were extracted by solid phase micro-extraction and dynamic head-space and then were analyzed by GC-MS, we use electroantennogram(EAG) and “Y” tube olfactomcter to test the response of Agrilus zanthoxylumi to the volatile components.We preliminarily study the mechanism of odor perception on Agrilus zanthoxylumi to the volatile components and know the the ultrastructure, type, number, distribution of sensilla on the antenna of Agrilus zanthoxylumi, and the differences of sensilla between male and female. Our research aimed at riching the study materials of Agrilus zanthoxylumi,supplement the morphology study of Agrilus zanthoxylumi. For the future chemical ecology prevention and the development of the plant source attractant or repellent to provide theoretical basis. The results of our study are as follows:1. β-caryophyllene(13.27%),trans-β-ocimene(16.82%), α-ocimene(16.42%) were included in the leaf of healthy Z. bungeanum, β-caryophyllene(13.27%), geranyl acetate(15.89%), Cubebene(17.27%) were mainly in the twig,(1R)-(+)-α-pinene(25.91%), β-caryophyllene(47.98%) mainly in phlome. When Z. bungeanum amaged by Agrilus zanthoxylumi the relative quantity of mycene,(1R)-(+)-limonene, linalyl acetate in Z. bungeanum were increased, the relative quantity of β-caryophyllene and geranyl acetate were decreased,and new volatiles were produced in phloem, like α-phellandrene.The component of volatiles in Z. bungeanum is almost the same which collectes by dynamic head-space and extracted by solid phase micro-extraction, ut the Content are different.besides, analysis showed amount of 1.3- diethyl benzene and 1.4- diethyl benzene is in both healthy and insect damaged Z. bungeanum.2. 11 kinds of volatiles can cause Agrilus zanthoxylumi EAG response, especially at the concentration of 100 μg/μL, but β-pinene at the concentration of 10 μg/μL. Myrene and(1R)-(+)-limonene caused female maximum EAG relative valule at the concentration of 10 μg/μL, while male at the concentration of 100 μg/μL. Except α-phellandrene, Agrilus zanthoxylumi adults have no significant gender differences in the EAG response among the volatiles.3.(1R)-(+)-α-pinene at the concentration of 1 μg/μL had abvious repellent effect both on male and female, β-pinene at the concentration of 0.1 μg/μL had abvious repellent effect on male, β-caryophyllene repellented female heavily at the concentration of 10 μg/μL(p<0.05). α-phellandrene at the concentration of 0.1 μg/μL, myrene at the concentration of 10 or 100 μg/μL,(1R)-(+)-limonene at the concentration of 0.1 or 1μg/μL can attract male. Linalyl acetate at the lower concentration(0.1 ~10μg/μL) can attract female, but at the concentration of 100 μg/μL exhibit repellent effect.4. Agrilus zanthoxylumi has the serrate antenna, which include 11 sections Secape, pedicel, flagellum(9 subsegments),There are 6 types(17 kinds) of sensilla on the antenna of male and female: including 3 kinds of sensilla trichodeas, 4 kinds of sensilla chaeticas, 7 kinds of sensilla basiconicas, 1 kind of sensilla campaniformia, 1 kind of sensilla gemmformium and 1 kind of unusual sensilla, besides B?hm bristles were only found on the antenna of male. The internal and external sides of the antenna have sensilla trichodeas, sensilla trichodeas and sensilla chaeticas were distributed over segmental venter and the back side. There is a fossa on the apex of every flagellomere from fourth to eleventh, and sensilla basiconicas were mainly distributed over it. Sensilla basiconicas cluster in the sunken place, where is beside the fossa, but the sensilla basiconicas cluster is not obvious on the tenth and eleventh flagellomeres.Our study tested the respond of Agrilus zanthoxylumi to the volitals of Z. bungeanum in both healthy and damaged,and chear the sensalia of Agrilus zanthoxylumi,which can provide the base for the futher research on olfactory function and the role of the volatile on host selection mechanism.
