C. perfringens is a gram-positive anaerobic rod-shaped bacterium, subiquitous in natural environment, intestinal tracts of human and animals,which is also one of the most common and important pathogens on human and animals. C.perfringens producing more than 17 toxins and enzymes, these toxins and enzyme are responsible for spectrum of diseases. Toxin producing strains have been divided into five distinct types of toxin(A to E) on the basis of their differential production of the four C. perfringens extracellular typingtoxins α, β, ε and ι.Plasmids as transferable gene elements, play a key role for toxins gene and antibiotic-resistance gene transferation of C. perfringens strains. Most of toxins gene of C. perfringens are encoded by plasmids, some plasmids carry both toxin genes and antibiotic-resistance genes. C. perfringens toxin gene and antibiotic-resistance gene transfer following with conjugative plasmids among different strains, spreaded toxins gene and antibiotic-resistance gene to different bacterias. In this study we compared and analysised three plasmids extraction methods of C. perfringens, and by method improving get more effective method. Study on plasmid extraction methods is make preparations for the future research.In order to understanding the C. perfringens 5 types of toxin’s distribution and antibiotic-resistance condition in different regions, we collected 704 samples from 5 provinces and 11 counties. Isolated 199 C. perfringens strains from 704 samples, the isolation rate is 28.5%. In 199 isolate C. perfringens strains, 187 isolated strains are type A, and 12 isolated strains are type C.Selecting 11 drugs that commonly used by vererinary treatment or feed additive for antibiotic sensitive experiment. Compared methods of Mahony(1986), Roberts(1986).and Hussain(1993) for plasmids extraction, result suggested that the method by Roberts(1986) get high purity of plasmid, but low concentration. So we based on this method and absorbed Hussain and Purnima(1993) experience, finally get improvement method and have better result.
