| Selenium is a trace element necessary for animal organism, plays a biological role by incorporating into the polypeptide chain of selenoprotein in the form of selenocysteine, particularly important in the biological function of the brain. Sep(O-phosphoserine)t RNA :Sec(selenocysteine)synthase(SepSecS) plays a crucial role in the synthesis and transformation of selenium acid, anda important role in biological function of selenium by synthesis and regulation of selenoproteins, however the role which SepSecS has in the regulation of selenium on selenoproteins transcriptome in chicken neurons remains is not clear. This test was fed deficiency and enriched selenium levels to chicken, detected the selenoproteins transcriptome level of different brain tissues in chicken, established the model of SepSec S gene silencing in chicken neuron using the technology of RNA interference, and observed the role of SepSecS in selenium regulated selenoproteins transcriptome. The results showed:1. Bioinformatic methods were used to analysis the structure and function of SepSecS, exhibiting that the protein invovled in the synthesis of selenocysteine by combinating with PLP, PO4, SEP; at some time, used a synthetic polypeptide with good immunogenicity, successed prepared the polyclonal antibody of rabbit against chicken SepSecS; used prepared polyclonal antibodies to detect the specific expression profile of SepSecS in the nervous system of chicken, thalamus> cerebrum> marrow> bulbus cinereus> cerebellum.2. Selenium regulated the expression of selenoprotein synthesis related factors in different brain regions of chicken, the expression level of SepSecS was significantly increased abundant selenium by in different brain regions of chicken; The expression of SepSecS protein and mRNA can be enhanced by selenium in vitro chicken embryo neurocytes.3. The content of selenium had a regulatory role in different brain regions of selenium protein expression profiling in chicken, we found the expression of Selt, Selk, Selu, Selw, Gpx3, Gpx4, Sepp1, Selo, Sel15, Selm, Gpx2 and Sepp2 were sensitived with the selenium content in the nervous system; Selenium had regulation role in selenoproteins transcriptome of chicken embryo neurons, the expression of Selt, Sel15, Selu, Gpx3, Selk, Selw, Gpx4, Sepp1 and Gpx2 were sensitive to the changes with the selenium content in chicken embryo neurocytes; Selenium content had an obvious role in regulated the selenoproteins transcriptomeof the cerebellum, thalamus and marrow.4. SepSecS silence model was successfully prepared using RNA interference technology in chicken; Dose-dependent effect of selenium content in selenoproteins expression in chicken neurons had disappeared, the expression of selenoproteins were significantly decreas ed, reflected that SepSecS had a key role in selenium regulated selenoproteins transcriptome. |
