Dietary selenium (Se) as an essential trace element has accepted considerable attention to the biological function and mechanism in the human beings and domestic animals, and through involved into selenoproteins which played the crucial role during oxidative stress, prevent cancer, redox and other cellular processes in vast majority tissues, affected various aspects of body, especially in the immune responses aspect, Although the mechanisms of this effective were not fully understood. Currently, almost25kinds of selenoproteins had known, which the main biological function is to antioxidant enzymes, but the biological function of selenoproteins still unclear. Chicken intestine not only is the main organ of selenium absorption, but also the intestinal mucosa was important defense organs in body. Selenium deficiency can cause the chick intestinal homeostasis broken, caused intestinal damage, but the effect and mechanism of selenium deficiency on the intestinal mucosal immune response remains unclear.In this experiment, total160day-old chicks were randomly divided into two groups. L group was fed the selenium content of selenium deficiency is0.033mg/kg basal diet; Control group was fed the selenium content of0.15mg/kg of the basal diet. Respectively,15d,25d,35d,45d and55d sacrificed collected duodenum, small intestine and rectum tissue. Using the microscopy to observe the histopathological changes in three different intestinal tissues. Detected the mRNA expression of23selenoproteins and immune-related cytokines in three different intestinal tissue by RT-PCR technology. Using the kit to detecte the antioxidant function of intestinal tissues. The purpose of this research was to discuss the effect and the mechanism of selenium deficiency on intestinal mucosal immune. The results showed that:1. The lack of a dietary selenium can cause intestinal tissue pathological chicks injury, intestinal mucosal bleeding, observed microscopic damage the integrity of the intestinal mucosa, inflammatory cell infiltration.2. Through23kinds of selenium in the detection of chicken intestinal protein expression levels of selenium are known, proven dietary selenium content can be lowered to reduce intestinal tissue mRNA expression levels of selenoproteins.3. Dietary selenium levels can affect different chick intestinal tissue immunoglobulin and cytokine mRNA expression levels, selenium can be proved by regulating the expression of cytokines in the intestinal mucosa of the intestinal mucosa and weakened immune function.4. Chicks selenium deficiency can cause intestinal tissue CAT activity increased significantly decreased, MDA and H2O2content GPx activity and GSH levels, suggesting that selenium can reduce the intestinal damage antioxidant capacity of the intestine caused.5. Selenium can affect the expression of heat shock proteins in chicken intestine proved to reduce heat shock protein function may be one mechanism of selenium cause intestinal injury.
