Storage proteins are synthesized and secreted into the hemolymph by fat body cells, they are recaptured by receptor-mediated by the fat body during larva-pupae metamorphosis for rebuilding the organization and structure of pupae. The honeybee is a holometabolic insect, it’s development undergo the stages of egg, larva, pupae and adult. It experienced the degradation of larva tissue and rebuild the organization and structure of pupae in the larva-pupae metamorphosis stage. Hexamerins are involved in this transversion. The foundational function of insect storage proteins is as a source of amino acids for development metamorphosis. Furthermore, it also play various roles in many biological functions such as metamorphosis, caste,-sex, oviposition, life span and immune.In order to probe the expression pattern of storage proteins in honey bees and their possible biological function, hexamerins in three castes honeybeeduring the metamorphosis were analyzed by using SDS-PAGE electrophoresis technology. The results revealed that few proteins can be detected with a little amount in larval haemolymph. In pupal stage, the quantity and variety of the proteins in haemolymph are booming, what is most distinct change is that four proteins with high level appeared in the haemolymph, consisted more than 80 percent of the total protein, with the bee larvae development, the four hexamerina gradually reduced, and undected after eclosion. Although expression profile of the four hexamerins is same during the development stages, but the content of a same hexamerin in haemolymph among worker, queen and drone is difference at the same development stage. It is more higher in the queen and drone than the worker haemolymph.RNAi has been widely applied in many kinds of genes function research. Synthetize enough pure dsRNA is the precondition in practical application,. In this study, hex110 dsRNA and hex70b dsRNA, were successfully synthetized by two methods, in vitro transcription and induced from E.coli. Especially, the establishment of the the system of induction synthesis technique by E.coli can be the basis for further study and subsequent application of honeybee related genes. Furthermore, we use the dsRNA synthetized by two methods to feed and inject worker, drone and queen bee respectively. The results show that the object hex110 and hex70b were not interfered seccessfully.In conclusion, hexamerins in different developmental stage of three honeybee castes were analyzed by using SDS-PAGE electrophoresis technology for evaluating the it’s function during the metamorphosis process. Moreover, dsRNA of hex110 and hex70b were synthetized by two methods, and then used for feeding and injecting worker, drone, queen honeybee respectively, for the purpose of evaluating the function of hexamerins in metamorphosisamong three castes of honeybee. The result will be as a reference for future related reaerch and application.
