Biocontrol And Metabolic Factors Analysis Of Bacillus Amyloliquefaciens TJ Strain

Bacillus amyloliquefaciens TJ strain as a biocontrol bacteria was isolated from rhizosphere soil of cucumber in 2008, we found the genome of TJ strain contain complete gene cluster of antibio-tics or promoting growth factors,the factors included such as Surfactin,Iturin,Fengycin,Sideropho-re,IAA,Macrolactin and so on.The three lipopeptid material family(Surfactins,Iturins,Fengycins) have different structures and chemical properties of the homologues or analogues which lead to the crude extraction or purifyc-ation has ability to resist fungi, bacteria or viruses, or have single resistance of anti fungal or bac-terial. Using of different fermentation, purification means to get substance which same as or similar to the standard to compare with the peak of HPLC(5μm,4.6×250mm).To detect and esti-mate the peak of Surfactin extract and Surfactin A standard in wavelength of 215 nm and 30% acetonitrile-water mobile phase conditions,found that the difference of peak time is less than 0.5 min. And TJ strains can produce three kinds of Surfactin substance during TLC purification process. In the wavelength of 214 nm, 45% methanol-water flow conditions, Iturin extract and Iturin A standard had the same peak time in 5.3min. At 214 nm and in 90% acetonitrilewater elution conditions, three analogue of Fengycin in extraction had same retention time with Fengycin standard at 5.35 min.IAA is a kind of trace signal molecules which has important significance in the regulation of plant growth and development. After detected the fermentation of cul-ture medium with L- tryptophan got that TJ strain can secret IAA in the metabolic processes. Siderophore can use of iron as a kind of bio-control factor to enhance bacteria itself and promote pla-nt ability of utilization of Fe. Using transparent circle of CAS plate method to verify the TJ strain can produce siderophore.Macrolactins have the potential to develop new drugs, but its biosynthetic output is very low, and Macrolactin family has so many homologous bring a lot of obstacles in separation and purify-cation of Macrolactins. Did not find the appropriate standard of Macrolactin for HPLC, therefore, using LC-MS analysis molecular weight to test existence of Macrolactin.At last,TJ strain could produce a kind of Macrolactin which molecular weight was about 420.
