This experiment take the Solanum torvum for the stock, the purple beauty eggplant variety of the scion grafting plant as the research object, and the same variety root eggplant is the comparison, to explore the change of physiological and biochemical substances in grafted eggplant healing process,the test materials were sampled with an 2-day interval after 2-14 day of treatment on the grafting union and wound own-rooted sampling,To study in grafted eggplant healing related enzyme activity, lignin content, phenolics content,.and seven different periods of the graft healing difference protein analyses. The finding indicated that:1.The results indicate that the activity of PAL and POD have Large variations during healing process compared with healing wound.In general,the activity of PPO of grafting union of eggplants and healing wound of own-rooted trees shows a little change,and both appears fist rised then declined.The activity of CAD and content of lignin of grafting union of eggplants and healing wound of own-rooted trees both performance rise tendency in the healing process,but healing wound is more dramatic.The activity of PPO.POD appears a peak after 12 days of grafting,it illustrates that the 12days is the critical point for union of grafting, rootstock and scion was basically completed in the 12 days.The rate of growth of grated seedling is slower than healing wound of own-rooted trces,the former was delayed 6 days than the later.2.Five kinds of phenolic substances in grafted eggplant healing process (procyanidins, chlorogenic acid, cinnamic acid, catechin, caffeic acid) content were determined by high performance liquid chromatography (HPLC),the results showed that the phenolic are peak value 12days after treatment at graft union, rootstock and scion of the docking healing was basically completed 12 days, proving that the increase of phenolics has no greater effect to the graft healing rate.The wound of own-rooted of five kinds of phenolics content change has been steady trend, that phenolics smaller variations improve healing rate. Procyanidins at the highest levels of the five kinds of phenols, but the graft union procyanidins and other physiological indexes showed no correlation, and the wound procyanidins and chlorogenic acid and catechin showed significant correlation; a significant correlation between Chlorogenic acid and various physiological indexes.3.Taking the grafting union of eggplant as research objects,testing the protein separation in grafting union by using 2-DE tecnique of two-dimensional eletrophoresis,comparing protein changes of seven different developmental stages in the healing process grafting union of eggplant.Determining 26 differential expression protein spots which expression differences are 2 and more than 2 times, and that repeatability is good. We conclude six changes of these protein spots:1 protein spot continued up-regulated; 3 protein spots continued down-regulated; 8 protein spots rose in the first stage, and then decreased; 3 protein spots decreased in the first stage, and then rose; 6 protein spots showed no regularity of dynamic; 5 protein spots disappeared after 14 days. In the healing process of the grafting union of eggplant,these different expressions of protein say that in the healing process of grafting union happens a series of changes in physiological and biochemical metabolic.The research of difference protein provides an experimental basis for idendifying protein mass spectrometry, studying the molecular mechanism and regulation of gene in the future.
