Broiler ascites syndrome (AS) is a kind of disease which is characterized by enlargement of right heart and large amount of ascites, which caused great economic losses to broiler breeding industry; The main cause of right heart hypertrophy is the rapid growth of broiler and the hypoxia of the body. It is an important link in the occurrence and development of AS in broiler chickens, Therefore, it is an effective way to reduce the incidence of AS in Broilers by exploring to feed safe and harmless substances for Broilers; As the most abundant free amino acid in cardiac cells, taurine has the function of antiapoptosis, regulation of Ca2+ homeostasis and protection of the heart.Objective:This experiment was passed by Low temperature to induce ascites syndrome in broilers, to investigate the inhibitory effect of taurine on broiler AS right ventricular hypertrophy, and provide a theoretical basis for the prevention and treatment of AS.Methods:In this experiment, a total of 180 one days old AA broilers were selected and randomly divided into three groups at 14 days old. Blank control group was fed in the environment of 20-22 ℃, low temperature model group was fed in the environment of 9-11 ℃ Taurine treatment group in the environment of 9-11℃ with feeding water containing 1% taurine. At the age of 21、28、35 and 42 age we selected randomly 10 broilers from each group, collecting and preparating of Venous blood and heart from broiler chickens. Venous blood for detecting the total number of red blood cells, red blood cells deposited, hemoglobin; We separated the right ventricular, to make the paraffin section, HE staining to observe the changes of the structure of the right ventricular muscle; We Separated the right ventricular, to prepare tissue homogenate,adopted tradioimmunoassay to mensurate contects of atrial natriuretic peptide (ANP), angiotensin angiotensin Ⅱ (Ang-Ⅱ) and endothelin-1 (ET-1) in right ventricular muscle; adopted Elisa to mensurate activies of Calpains(Calpain) and Cytochrome-C (Cyt-C); meanwhile, Statistics incidence of ascites in the whole trial cycle and the ascites heart index (AHI) at different stages.The results were as follows:1.Taurine could significantly reduce the incidence of ascites and ascites heart index of AS broilers induced by low temperature.2.Taurine could improve pathological changes of the right ventricular tissue of AS broilers induced by low temperature.3.Taurine could reduce the content of RBC,PCV, MCV and HGB in the blood of AS broilers induced by low temperature,then reduced blood viscosity, reduced the right heart load, resistanced to the right heart hypertrophy.4.Taurine could significantly reduce the content of ANP, Ang-and ET-1 in the right ventricular tissue of AS broilers induced by low temperature, so as to reduced the load of the right ventricle and the right ventricular hypertrophy.5.Taurine could reduce the activity of Cyt-C and Calpain in the right ventricular tissue of AS broilers induced by low temperature, prevented right cardiac hypertrophy by reducing apoptosis of cardiomyocytes induced by Calpain and Cyt-C.In conclusion, Taurine could improve right ventricular muscle tissue structure by decreasing the viscosity of blood, the content of ANP, ET-1 and Ang-Ⅱ and apoptosis related factor Calpain and Cyt-C activity in cardiac cells of AS broilers induced by low tempereture, resisted the right heart hypertrophy, so as to prevent the broiler ascites syndrome.
