Microstructure Observation Of Different Organs And Tissues Of Broiler Ascites Syndrome And Analysis Of Differential Expression Of HIF-1α And VEGF

Abstract/Summary:
Ascites syndrome(AS)of broiler chickens is a common disease in large-scale broiler farms,which occurs worldwide and causes great harm to the healthy development of broiler breeding industry.In this study,10-week-old AS broilers from a broiler farm in the suburb of Aksu,Xinjiang province were selected AS the research subjects to investigate the morbidity of AS broilers.Pathological observation and analysis of lung,heart and liver of AS broilers were conducted by HE staining,Masson trichroor staining and Van Gieson staining.Meanwhile,molecular biological methods such as immunohistochemistry,Real-time fluorescence quantitative PCR and western blot were used.The expression and distribution of Hypoxia inducible factor-1ɑ(HIF-1ɑ)and Vascular endothelial growth factor(VEGF)in lung,heart and liver of AS broilers were detected.To analyze the regulatory roles of HIF-1ɑ and VEGF in lung,liver and heart of AS broilers during the pathogenesis,in order to provide reference for the prevention and treatment of AS in this area.The histological changes of lung,heart and liver in AS broilers were observed.Compared with healthy broilers,pulmonary artery vessels of AS broilers had obvious remodeling phenomenon,and the boundary between intima,media and adventitia was not clear after remodeling,and intima thickened significantly.The normal structure of liver tissue was completely lost,liver cords were loosely arranged,and some normal liver cells were replaced by inflammatory cells and fibroblasts.Cardiac tissue fibrosis,irregular arrangement of myocardial fibers,inflammatory cells increase;AS broiler lung collagen fiber content increased significantly,but heart and liver collagen fiber did not change significantly.qRT-PCR results showed that compared with healthy broilers,HIF-1ɑ expression was significantly different in liver and lung(P<0.01),but not in heart(P>0.05).VEGF expression was significantly different in liver of AS broilers(P<0.05),extremely significant in lung(P<0.01),but not in heart(P>0.05).Western-blot results showed that the expression of HIF-1ɑ protein in liver,heart and lung of AS broilers was significantly higher than that of healthy broilers(P<0.01),and the expression level of heart was higher than that of liver and lung.The protein content of VEGF was basically the same AS HIF-1ɑ,and the expression of VEGF in liver,heart and lung of AS broilers was extremely significantly higher than that of healthy broilers(P<0.01),and the expression level in heart was higher than that in liver and lung.Immunohistochemical results showed that compared with healthy broilers,HIF-1ɑ expression was significantly different in lung,heart and liver of AS broilers(P<0.05),and the positive signal of liver HIF-1ɑ was the strongest.The expression of VEGF in heart and liver of AS broilers was significantly different(P<0.05),but not in lung(P>0.05),and the positive signal of VEGF in heart was the strongest.The results showed that AS occurred in a broiler farm in aksu suburb,and the incidence of AS increased significantly in winter.After the onset of the disease,pathological changes occurred in the tissue structure of lung,heart and liver of AS broilers,and the distribution of collagen fibers around pulmonary artery increased,but the collagen fibers in liver and heart had no significant changes.Compared with healthy broilers,HIF-1ɑ was significantly expressed in the lungs,heart and liver of AS broilers,while VEGF was significantly expressed in the heart and liver of AS broilers,but not in the lung.The m RNA expression of HIF-1ɑ and VEGF in lung and liver of AS broilers was significantly different from that of healthy broilers,but not in heart.The expression of HIF-1ɑ and VEGF proteins in lung,liver and heart of AS broilers is significantly different from that of healthy broilers,and the expression tends to be basically the same.
