| Taurine is widely distributed in various tissue organs of animals and human bodies, and is one of the most abundant free amino acids in organism. Taurine plays an important role in regulating and maintaining normal physiological functions, including anti-oxidation, biomembrane stabilization, cell protection, osmoregulation, calcium homeostasis regulation, and anti-inflammation. It is generally accepted that taurine is mainly synthesized in the liver via cysteine dioxygenase (CDO)-cysteine sulfinic acid decarboxylase (CSD) pathaway, and transferred into cells by the taurine transporter (TAUT) on cytomembrane to exert its biological effect. But so far, there is no detailed information about the distribution and synthesis of taurine in dog’s tissue organs, which limits the function research and application of taurine in dogs. In this experiment, the contents of taurine in canis lupus familian’s liver, hypothalamus, pituitary, thyroid, adrenal, testicle and ovary, and the expression of mRNA and protein levels of key enzymes (CDO and CSD) as well as taurine transporter TUAT were detected through enzyme linked immunosorbent assay (ELISA), real-time fluorescence quantification PCR method, and Western blot method. The results will lay a foundation for further study of taurine’s role in canis lupus familian’s endocrine system and its application in canis lupus familians.The result demonstrates that:taurine exists in canis lupus familian’s liver, hypothalamus, pituitary, thyroid, adrenal, testicle, ovary and serum, and mostly in the pituitary. CDO, CSD and TUAT are expressed in canis lupus familian’s liver, hypothalamus, pituitary, thyroid, adrenal, testicle, and ovary. The expression level of CDO mRNA in descending order is: testicle, adrenal, liver, ovary, thyroid, pituitary, hypothalamus; the expression level of CSD mRNA in descending order is:testicle, ovary, thyroid, liver, adrenal, pituitary, and hypothalamus; the expression level of TAUT mRNA in descending order is:ovary, testicle, liver, adrenal, pituitary, thyroid, and hypothalamus.The results indicated that taurine can be biosynthesized through CDO-CSD pathway in canis lupus familian’s liver, hypothalamus, pituitary, thyroid, adrenal, testicle, and ovary. However, the content of taurine biosynthesized can not meet the need of organs function, and must obtain from the blood by taurine transporter. Taurine plays an important role in the function of liver, hypothalamus, pituitary, thyroid, adrenal, testicle and ovary, but the exact function needs further investigation. |
