As the biologically active ingredient of medicinal plants Macleaya cordata, sanguinarine has significant antibacterial activity. Due to the significant antibacterial activity, its pharmacological efficacy research carried out continuously, but the pharmacokinetic studies of sanguinarine reported rarely. To further clarify the pharmacokinetics in vivo as a veterinary drug, and provide experimental data for safety evaluation of sanguinarine, the pharmacokineticd and tissue distribution of sanguinarine in chicken were stuied. In this paper, a simultaneous quantitative determination of sanguinarine, chelerythrine, dihydrosanguinarine and dihydrochelerythrine in chicken by HPLC-MS/MS method was established, the pharmacokinetic of sanguinarine in chick plasma were investigated, and the tissue distribution of sanguinarine, chelerythrine and their metabolite in vivo were determined also.1. The broiler tissues samples were prepared to test solution by the process of extraction and separation, and determined using high performance liquid chromatography electrospray ionization-triple quadrupole tandem mass spectrometry. By optimizing the LC-MS/MS determination conditions, a simultaneous quantitative determination of sanguinarine, dihydrosanguinarine, chelerythrine and dihydrochelerythrine in chicken by HPLC-MS/MS method was established. The method were validated in terms of specificity, linearity, precision, accuracy and stability, the results show that the method has good specificity, high accuracy, good precision and high sensitivity. Therefore, this method is suitable to measure the concentration of sanguinarine, dihydrosanguinarine, chelerythrine and dihydrochelerythrine in vivo tissues.2. To investigate the pharmacokinetics of sanguinarine and dihydrosanguinarine, six chicks were intragastric administrated a single oral doses of 30,15 and 10 mg.kg’1 sanguinarine. HPLC-MS/ms method was used for the quantification determination of sanguinarine and dihydrosanguinarine in plasma samples. The pharmacokinetic parameters were calculated by DAS3.0 pharmacokinetic program.The results showed that SG and DHSG had a short Tmax with 0.3±0.30h and 0.25±0.12h after a single dose of SG administration, which implied that SG has high adsorption in chick plasma. The Cmax of SG was smaller than that of DHSG. It meant SG could be reduced to DHSG in chick.3. Selected the one week old broiler as experiment subject, the chicks were divided into groups, one group was fed the fodder containing Sangrovit as additive(30mg Sangrovit added into 1.0 kg fodder) for100 days. The other goup was fed the normal fodder for 100 days. When expiration, the chicks were sacrificed, and the chick tissues were collected. The drug residues of sanguinarine, dihydrosanguinarine, chelerythrine and dihydrochelerythrine in chick tissues were quantitative determined using the established HPLC-MS/MS methods. The results showed that the content of sanguinarine, dihydrogsanguinarine, chelerythrine and dihydrochelerythrine in chicken tissue is existed but very low, However, after stop feding the fodder containing Sangrovit as additive for seven days, the four kind of alkaloids can not be detected in chick tissue.
